摘  要:目的:探索葛根及其配伍对骨骼肌病理形态及其线粒体ATP酶的影响。方法:将160只健康大鼠随机平均分为8组,分别为正常组、模型组、强的松组、葛根组、葛芪组、葛参组、葛妙组和葛芪参妙组,用鼠源性乙酰胆碱受体97~116肽段作为免疫原,多点皮下注射以造模,成模后分别使用强的松和各组中药进行连续灌胃4周,然后取材,分别观察大鼠治疗前后骨骼肌病理形态的改变及其骨骼肌线粒体ATP酶的变化。结果:相比正常组,模型组重症肌无力大鼠骨骼肌线粒体的ATP酶活性明显降低,其Na^+-K^+-ATPase,Mg^2+-ATPase,Ca^2+-ATPase,Mg^2+-Ca^2+-ATPase活性明显低于正常组大鼠(P<0.05);治疗后,强的松组和各中药治疗组大鼠的ATP酶活性均有所恢复(P<0.05),以葛芪参妙组的恢复最为明显(P<0.01)。结论:葛根及其配伍组合可能通过提高骨骼肌线粒体ATP酶活性以改善骨骼肌病理形态,从而治疗重症肌无力。Objective:To explore the effect of Pueraria lobata and its compatibility on skeletal muscle pathology and mitochondrial ATPase.Methods:160 healthy rats were randomly divided into 8 groups:normal group,model group,prednisone group,Gegen group,Geqi group,Geshen group,Gemiao group,Geqi Shenmiao group,Rat-derived acetylcholine receptor 97~116 peptide segment was used as immunogen,and multi-point subcutaneous injection was used to establish the model.Results:Compared with the normal group,the ATPase activity of skeletal muscle mitochondria in the model group was significantly lower,and the activities of Na^+-K^+-ATPase,Mg^2+-ATPase,Ca^2+-ATPase,Mg^2+-Ca^2+-ATPase were significantly lower than those in the normal group(P<0.05);after treatment,the ATPase activity of the prednisone group and the rats in the treatment group of Chinese herbs all recovered(P<0.05),especially in the Geqi Shenmiao group(P<0.05).Conclusion:Gegen and its compatible group may improve the pathological morphology of skeletal muscle by improving the activity of mitochondrial ATPase in skeletal muscle,thus treating myasthenia gravis.
