摘 要:目的研究基质相互作用分子1(STIM1)在高糖诱导系膜细胞损伤中的作用及机制。方法以人肾小球系膜细胞作为研究对象,实验分为正常组(N组),高糖(high glucose)作用12小时及24小时组(HG12h及HG24h组),应用Real-time PCR及Western-blot方法分别检测在不同作用时间下高糖引起系膜细胞STIM1 mRNA及蛋白表达变化,通过激光共聚焦实验观察在高糖诱导下系膜细胞内钙变化,以及应用Tunel实验检测系膜细胞凋亡情况,观察应用STIM1siRNA基因转染对系膜细胞内钙及凋亡的影响。结果实验显示,HG12h组与N组比较STIM1mRNA含量及蛋白表达均明显增加(P<0.05),HG24h组与N组比较STIM1mRNA含量及蛋白表达显著增加(分别为P<0.01及P<0.05);激光共聚焦结果表明,HG12h组及HG24h组与N组比较,系膜细胞内钙明显升高(P<0.05);Tunel实验显示,HG12h组及HG24h组与N组比较,系膜细胞凋亡增加(P<0.01);STIM1siRNA基因转染组与未转染组相比系膜细胞内钙明显减少(P<0.05),细胞凋亡减轻(P<0.05)。结论高糖可以诱导系膜细胞内钙变化,细胞发生凋亡,STIM1在其中起着重要作用。Objective To investigate the role and mechanism of stromal interaction molecule 1(STIM1)in mesangial cells injury induced by high glucose.Methods Human glomerular mesangial cells were taken as study subjects.The experiment was divided into normal group(N group),high glucose for 12 hours and 24 hours group(HG12 h and HG24 h group).Real-time PCR and Western-blot were used to detect the changes in STIM1 mRNA and protein expression of mesangial cells induced by high glucose at different time points.The changes of calcium in mesangial cells induced by high glucose were observed by laser confocal assay,and the apoptosis of mesangial cells was detected by Tunel assay.The effects of STIM1 siRNA gene transfection on calcium and apoptosis in mesangial cells were observed.Results The experiments showed that STIM1 mRNA and protein were significantly increased in HG12 h group compared with that of N group(P<0.05).STIM1 mRNA and protein were significantly increased in HG24 h group compared with that of N group(P<0.01 and P<0.05,respectively).The results of laser confocal imaging showed that calcium in mesangial cells was significantly increased in HG12 h group,HG24 h group compared with N group(P<0.05).Tunel assay showed that the apoptosis of mesangial cells increased in HG12 h group,HG24 h group compared with N group(P<0.01).The intracellular calcium and apoptosis significantly reduced in STIM1 siRNA gene transfection group compared with non-transfection group(P<0.05).Conclusion High glucose can induce intracellular calcium changes and apoptosis of mesangial cells,in which STIM1 plays an important role.
