摘  要:目的研究胸腺素β4(Tβ4)联合外源性碱性成纤维细胞生长因子(bFGF)对深Ⅱ度烫伤大鼠创面愈合的影响及其机制。方法将40只深Ⅱ度烫伤大鼠随机分为正常对照组(外涂生理盐水)、Tβ4组(外涂6μg Tβ4)、bFGF组(外涂4μg bFGF)及Tβ4+bFGF组(外涂6μg Tβ4+4μg bFGF),每组各10只。给药第3、7、10、14 d对创面进行拍照并取创面皮肤标本,计算各组大鼠创面愈合率,利用RT-PCR检测TGF-β1 mRNA及VEGF mRNA表达,Western blot检测VEGF、Caspase-3及NF-κB蛋白表达。给药后第1、3、6、12、24及48 h,取各组大鼠创面皮肤标本,采用免疫组织化学法检测IL-10及TNF-α阳性表达。结果给药后各时间点,Tβ4+bFGF组创面愈合率均高于其他各组(P<0.05),TGF-β1 mRNA表达量低于其他各组(P<0.05),而VEGF mRNA表达量及VEGF、Caspase-3及NF-κB蛋白表达量高于其他各组(P<0.05)。Tβ4+bFGF组IL-10及TNF-α阳性细胞数先增加后降低,12 h到达峰值,12 h前高于Tβ4组及bFGF组,12 h后低于Tβ4组及bFGF组(P<0.05)。结论Tβ4联合外源bFGF治疗深Ⅱ度烫伤能通过改善炎症反应,加速坏死组织细胞凋亡,促进新生血管生成加速创面愈合,具有很好的疗效。Objective To explore the effects of thymosin β4(Tβ4)combined with exogenous basic fibroblast growth factor(bFGF)on wound healing in rats with deep Ⅱ degree scald.Methods 40 molding rats with deep Ⅱ degree scald were randomly divided into normal control group(external of saline),Tβ4 group(external application of 6μg Tβ4),bFGF group(external application of 4μg bFGF)and Tβ4+bFGF group(external application of 6μg Tβ4+4μg bFGF).On the 3rd,7th,10th and 14th d of administration,the wounds were photographed and the wound skin samples were taken.The wound healing rate in each group was calculated.The expression levels of TGF-β1 mRNA and VEGF mRNA were detected by RT-PCR.The protein expression levels of VEGF,Caspase-3 and NF-κB were detected by Western blot.On the 1^st,3^rd,6^th,12^th,24^th and 48^th h after administration,the wound skin samples of each group were taken,and the positive expression levels of IL-10 and TNF-α were detected by immunohistochemistry.Results At each time point after administration,the wound healing rate in Tβ4+bFGF group was higher than that in the other groups(P<0.05),and the expression level of TGF-β1 mRNA was lower than that in the other groups(P<0.05),while the expression level of VEGF mRNA and the protein expression levels of VEGF,Caspase-3 and NF-κB were higher than those in the other groups(P<0.05).The quantities of IL-10 and TNF-α positive cells in Tβ4+bFGF group were increased first and then decreased,and peaked at 12 h,and the quantities were higher than those in Tβ4 group and bFGF group before 12 h and were lower than those in Tβ4 group and bFGF group after 12 h(P<0.05).Conclusion Tβ4 combined with exogenous bFGF can improve the inflammatory response,accelerate the apoptosis of necrotic tissue cells,promote the angiogenesis and accelerate wound healing for deep Ⅱ degree scald.
