摘  要:目的探讨胡椒碱对N-甲基-D-天冬氨酸(NMDA)诱导的大鼠嗜铬细胞瘤PC12细胞毒性的保护作用。方法将体外培养的高分化PC12细胞分为正常对照组、NMDA 300μmol/L组(模型组)、胡椒碱0.1μmol/L和1μmol/L处理组。检测各组PC12细胞的存活率和细胞内活性氧(ROS)水平、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性以及脑源性神经营养因子(BDNF)蛋白和mRNA的表达水平。结果与正常对照组相比,模型组细胞存活率、SOD活性、BDNF蛋白和mRNA表达降低,细胞内ROS水平和MDA含量升高(P<0.01)。与模型组相比,胡椒碱1μmol/L处理组细胞存活率、SOD活性、BDNF蛋白和mRNA表达升高,细胞内ROS水平和MDA含量降低(P<0.01)。结论胡椒碱对NMDA诱导的PC12细胞毒性具有保护作用,至少部分是通过抑制氧化应激和上调BDNF表达而实现的。这种神经保护作用可能是胡椒碱体内抗抑郁作用的途径之一。Objective To study the protective effect of piperine on N-methyl-D-aspartate(NMDA)-induced neurotoxicity in cultured rat pheochromocytoma(PC12) cells.Methods The high differentiated PC12 cells cultured in vitro were divided into four groups.PC12 cells of group A were incubated with NMDA 300 μmol/L,which of group B were incubated with NMDA 300 μmol/L plus piperine 0.1 μmol/L,of group C were incubated with NMDA 300 μmol/L plus piperine 1 μmol/L and of group D were incubated with DMEM medium as the controls.The relative cell viability,intracellular reactive oxygen species(ROS) level,malondialdehyde(MDA) content,superoxide dismutase(SOD) activity and protein and mRNA expression levels of brain-derived neurotrophic factor(BDNF) in four groups were detected.Results Compared with group D,the relative cell viability,intracellular SOD activity and protein and mRNA expression levels of BDNF were decreased,while intracellular ROS level and MDA content were increased in group A(P<0.01).Compared with group A,the relative cell viability,intracellular SOD activity and protein and mRNA expression levels of BDNF were increased,while intracellular ROS level and MDA content were decreased in group C(P<0.01).Conclusion Piperine has a neuroprotective effect on NMDA-induced neurotoxicity in PC12 cells,at least in part,via the inhibition of oxidative stress and the upregulation of BDNF expression.This neuroprotective effect may be one of the action pathways accounting for the in vivo antidepressant activity of piperine.
