摘  要:目的:测定大血藤中总蒽醌的含量;利用体外抗氧化模型,测定比较大黄素、大黄酚体外抗氧化能力的强弱,探讨其抗氧化能力产生的量效关系,为寻找酚酸类的抗氧化活性能力提供依据。方法:采用紫外分光光度法测定大血藤总蒽醌的含量;采用经典Fenton反应,根据公式计算对程自由基(•OH)的清除率,建立量效关系曲线,通过量效曲线算出清除率为50%时样品的浓度(IC50)值。结果:测得大血藤中总蒽醌的平均含量为0.00284%;两种酚性化合物和维生素C清除除羟自由基的IC50值大小顺序为:维生素C>大黄素>大黄酚。结论:本研究所用方法简便、准确,可作为大血藤中总蒽醌的含量测定方法;大血藤有效成分大黄素、大黄酚具有明显的体外抗氧化活性。Objective:To determine the total anthraquinone in sargentodoxae caulis.In vitro antioxidant models has been used to determine and compare the emodin's strength of antioxidant capacity in vitro to investigate the dose-effect relationship of antioxidant capacity,providing evidence for finding the antioxidant activity of phenolic acids.Methods:Ultraviolet spectrophotometry was used to determine the total anthraquinone in sargentodoxae caulis,the classic Fenton reaction was adopted to caculate the hydroxyl radical(OH)clearance rate according to the formula so as to establish the dose-effect curves,and through which,the concentration(IC50)value of the sample with a clearance rate of 50%was calculated.Results:The IC50 average content of total anthraquinone in sargentodoxae caulis was 0.00284%;the order of IC50value in the two phenolic compounds and vitamin C scavenging hydroxyl radicals was:Vitamin C>emodin>chrysophanol.Conclusion:This method is simple and precise,which can be used as the method to determine the total anthraquinone in sargentodoxae caulis.And emodin and rheinol,the active component of sargentodoxae caulis,have obvious antioxidant activity.
