摘 要:目的探讨黄苓苷与头孢哌酮-舒巴坦钠(cefoperazone-sulbactam sodium,SCF)联用对产ESBLs大肠埃希菌的作用方式。方法选取头孢哌酮-舒巴坦钠敏感和耐药的ESBLs阳性大肠埃希菌菌株各一株。黄苓苷和头孢哌酮-舒巴坦钠分别配置成母液,灭菌蒸馏水进行系列稀释,添加M-H培养基,设置不同处理组和对照组。以600nm处的光密度(optical density,OD)变化(OD20-OD0)评价不同处理对5×10^6 CFU/ml实验菌液的影响,标准菌株做平行对照。结果商品化的黄苓苷溶解度降低,杀菌能力丧失,抑菌能力存疑;与SCF的联用效果有菌株和浓度依赖性。结论商品化的黄苓苷没有表现出抗菌能力,不能直接用作抑菌药品,实验不支持其与SCF的联用;加碱反应能否恢复黄苓苷的天然属性需要进一步的研究。Objective:To investigate the effects of baicalin combined with cefoperazone-sulbactam sodium(SCF)on the ESBLs-producing escherichia coli(e.coli).Methods:Two positive e.coli strains that are sensitive and resistant to SCF were selected and the standard strains were used as parallel controls.Commercialized baicalin and SCF were respectively prepared as the mother liquor,serially diluted with sterile distilled water,and M-H medium was added to set different treatment and control groups.The effects of different treatments on 5×106 CFU/mL bacteria was evaluated by optical density change(OD20-OD0)at 600 nm.Results:The solubility of commercialized baicalin was reduced and the bactericidal ability was lost in its concentrating and refining process,and the antibacterial ability was doubtful too.And the synergistic effect with SCF was dependent on the strain and concentration.Conclusion:Some commercially available baicalin does not exhibit antibacterial ability and cannot be directly used as a bacteriostatic drug.Besides,the experiment does not support its combination with SCF.Whether the alkali reaction can restore the natural properties of baicalin requires further researches.
