摘 要:【目的】为评估施氏鲜和西伯利亚歸后备亲鱼群体的遗传多样性,为歸鱼的遗传选育提供参考。【方法】以施氏鳏和西伯利亚歸亲鱼群体为研究对象,进行了线粒体控制区(D-loop)全序列和细胞色素6(Cyt 6)基因序列对比分析,探讨2标记对歸鱼遗传多样性分析结果的差异与关系。【结果】西伯利亚歸和施氏MCyti和D-loop的A+T含量均高于G+C含量,碱基组成具有偏倚性。Cyt 6基因在西伯利亚歸中定义单倍型3个,多态性位点5个,单倍型多样性0.151,核甘酸多样性0.00036;在施氏歸中定义单倍型4个,多态性位点4个,单倍型多样性0.612,核昔酸多样性0.00114o D-loop在西伯利亚歸中定义单倍型3个,多态性位点2个,单倍型多样性0.385,核昔酸多样性0.00040;在施氏歸中定义单倍型5个,多态性位点55个,单倍型多样性0.707,核昔酸多样性0.02242o【结论】西伯利亚歸和施氏歸亲鱼群体的遗传多样性匮乏,尤其在利用西伯利亚鳏亲鱼进行繁殖育苗时要注意近交的不利影响。[Objective]The study aimed to evaluate genetic diversity of Acipenser baerii and A.schrenkii populations,and offer a reference for genetic breeding.[Method]In this paper the authors took the reserved parent fish broodstocks of A.baerii and A.schrenkii populations as the research object,by comparison of imtochondrial DNA cytochrome b(Cyt 6)gene and control region(D-loop)sequences to investigate the relation and difference genetic diversity relation of A.Baerii and A.Schrenkii with the two molecular markers.[Result]Whatever A.baerii or A.schrenkii,the content of A+T base pairs were greater than that of the G+C base pairs in both Cyt b gene and D-loop sequences,indicating that the base composition was biased.Three haplotypes were defined and five polymorphic sites(S)were detected in Cyt b gene of 4.baerii,while its haplotype diversity(H&)was 0.151 and nucleotide diversity(tf)was 0.00036.Four haplotypes were defined and four polymorphic sites were detected in Cyt b gene sequences of A.fiaezii,while its haplotype diversity was 0.612 and nucleotide diversity was 0.00114.Three haplotypes were defined and two polymorphic sites were detected in D-loop sequences of A.baerii,while its haplotype diversity was 0.385 and nucleotide diversity was 0.00040;Five haplotypes were defined and 55 polymorphic sites were detected in D-loop sequences of A.baerii,while its haplotype diversity was 0.707 and nucleotide diversity was 0.02242.[Conclusion]The results indicated that a marked low genetic diversity in reserved parent fish broodstocks,and more attention should be paid to avoiding the inbred risk in sturgeon fry production.
