摘 要:目的探讨4-b-佛波醇-12-豆蔻酸-13-乙酸酯(PMA)对未分化和分化的MO3.13少突胶质细胞铁相关蛋白表达的影响。方法将MO3.13细胞分为对照组和PMA组。对照组用基础培养液进行细胞培养,PMA组的培养液内加入终浓度为100 nmol/L的PMA,每3 d换1次液。培养7 d后,采用蛋白质免疫印迹实验检测两组细胞转铁蛋白受体1(TfR1)和铁调节蛋白1(IRP1)的表达水平。结果与对照组相比,PMA组细胞TfR1和IRP1的表达水平均显著降低(t=3.002、2.654,P<0.05)。结论100 nmol/L的PMA可引起少突胶质细胞TfR1和IRP1的表达降低,这种变化可能与IRP1的调节有关。Objective To investigate the effect of 4-b-phorbol-12-myristate-13-acetate(PMA)on the expression of iron-related proteins in undifferentiated and differentiated MO3.13 oligodendrocytes.Methods MO3.13 oligodendrocytes were divi-ded into control group and PMA group.The oligodendrocytes in the control group were cultured with a basic culture medium,and those in the PMA group were cultured with a culture medium containing PMA at a final concentration of 100 nmol/L;the medium was changed every 3 days.After 7 days of culture,Western blotting was used to measure the expression of transferrin receptor 1(TfR1)and iron regulatory protein 1(IRP1).Results Compared with the control group,the PMA group had significant reductions in the expression of TfR1 and IRP1(t=3.002 and 2.654,P<0.05).Conclusion PMA at a concentration of 100 nmol/L can reduce the expression of TfR1 and IRP1 in oligodendrocytes,which may be associated with IRP1 regulation.
