摘 要:目的探讨miR-33表达与炎症反应的相关性。方法细胞培养箱中培养人单核细胞(THP-1)48~72 h后传代,传至3代,加入100 ng/ml佛波醇(PMA)诱导24 h分化成为巨噬细胞。采用实时荧光定量PCR技术检测不同浓度LPS(0、1、10、100 ng/ml)以及10 ng/ml LPS不同刺激时间(0、24、36、48 h)THP-1巨噬细胞miR-33表达;另采用ELISA法检测THP-1巨噬细胞中TNF-α、IL-6和MCP-1表达量,分析miR-33表达与炎症反应的相关性。结果随着LPS浓度(0、1、10、100 ng/ml)增加,miR-33表达、TNF-α、IL-6和MCP-1水平逐渐增加,不同浓度组miR-33表达、TNF-α、IL-6和MCP-水平比较,差异有统计学意义(P<0.05)。随着LPS刺激时间(0、24、36 h)延长,miR-33表达逐渐上调、TNF-α、IL-6和MCP-1水平逐渐增加,36 h达到高峰,48 h逐渐下降到正常,不同时间点miR-33表达、TNF-α、IL-6和MCP-1水平比较,差异有统计学意义(P<0.05)。Pearson相关性分析显示,miR-33表达与TNF-α、IL-6和MCP-1呈正相关(P<0.05)。结论THP-1细胞中miR-33表达与其炎症反应具有一定正相关性。Objective To investigate the correlation between miR-33 expression and inflammatory response.Methods Human mononuclear cells(THP-1)were cultured in a cell culture incubator for 48 to 72 h and then passaged to passage 3,and 100 ng/ml phorbol(PMA)was added to induce macrophage differentiation for 24 h.Detection of THP-1 macrophages miR-33 at different concentrations of LPS(0,1,10,100 ng/ml)and 10 ng/ml LPS at different stimulation times(0,24,36,48 h)using real-time PCR expression;ELISA was used to detect the expression of TNF-α,IL-6 and MCP-1 in THP-1 macrophages,and the correlation between miR-33 expression and inflammatory response was analyzed.Results With the increase of LPS concentration(0,1,10,100 ng/ml),the expression of miR-33,TNF-α,IL-6 and MCP-1 levels gradually increased.There were statistically significant differences in miR-33 expression,TNF-α,IL-6 and MCP-levels in different concentration groups(P<0.05).With the extension of LPS stimulation time(0,24,36 h),miR-33 expression gradually increased,TNF-α,IL-6 and MCP-1 levels gradually increased,peaked at 36 hours,and gradually decreased to normal at 48 h.There were statistically significant differences in miR-33 expression,TNF-α,IL-6 and MCP-1 levels at time points(P<0.05).Pearson correlation analysis showed that miR-33 expression was positively correlated with TNF-α,IL-6 and MCP-1(P<0.05).Conclusion The expression of miR-33 in THP-1 cells has a positive correlation with its inflammatory response.
