摘 要:目的探讨麒麟丸对奥硝唑(ORN)造模所致弱精子症大鼠模型精子活力的保护作用及可能调控机制。方法采用400mg/(kg·d)ORN灌胃28d,复制弱精子症大鼠模型。将大鼠随机分为空白组、模型组、麒麟丸组和左卡尼汀组,每组10只,连续给药4周后,处死大鼠,检测大鼠附睾的精子浓度、总活力,附睾L-肉碱的含量,附睾OCTN2 m RNA的表达,并观察大鼠睾丸组织HE染色切片。结果与模型组相比,麒麟丸可明显提高精子总活力(P<0.01)、提高附睾L-肉碱的含量(P<0.01)、增加附睾OCTN2 m RNA的表达(P<0.01),并能改善奥硝唑造模导致的睾丸损伤病理。结论本实验发现,麒麟丸具有明显提高弱精子症大鼠模型精子活力的作用,其作用机制可能与其提高附睾L-肉碱的含量和OCTN2 m RNA的表达有关,更详细的作用机制有待进一步研究。Objective To investigate the protective effect and possible regulatory mechanism of Qi Lin Pill on sperm motility in ornidazole-induced asthenospermia rats.Methods Asthenospermia rat models were established by intragastric administration of ornidazole 400 mg/(kg·d)for 28 days.The established rat model were randomly divided into four groups including the blank group,the model group,Qi Lin Pill group and levocarnitine group(n=10).After continuous administration for 4 weeks,the concentration and motility of epididymis sperm,the content of L-carnitine in epididymis and the expression of OCTN2 m RNA in epididymis were measured,and histopathology of testis was observed.Results Compared with the model group,Qi Lin Pill could significantly promote the percentage of PR+NP grade spermatozoa(P<0.01),the content of L-carnitine in epididymis(P<0.01)and m RNA expression of OCTN2 in epididymis(P<0.01).Simultaneously,it improved ornidazole-caused testicular injury.Conclusion Qi Lin Pill can significantly enhance the sperm motility of asthenospermia rats,which may be associated with the increase of L-carnitine content and OCTN2 expression in epididymis.The detailed mechanism needs to be further investiagted in the future.
