摘  要:目的研究β-蜕皮甾酮(β-Ecd)介导β-catenin信号转导通路对膝骨关节炎小鼠的作用及其机制。方法按体重将3月龄雄性C57小鼠随机分为3组:假手术组、模型组和实验组,每组10只。用手术方法制备骨关节炎小鼠模型。于造模2个月后,实验组皮下注射β-Ecd 5 mg·kg^-1,假手术组和模型组给予0.9%NaCl,每天2次,连续2个月。以实时定量-聚合酶链反应检测关节软骨组织基质金属蛋白酶13(MMP13)、X形胶原(Col X)和骨钙素(OC)基因表达水平。结果假手术组、模型组和实验组的MMP13基因相对表达量分别为1.00±0.02,1.87±0.03和1.22±0.04;这3组的Col X相对表达量分别为1.01±0.04,2.92±0.21和1.61±0.10;这3组的OC相对表达量分别为1.00±0.05,1.72±0.03和1.19±0.02;模型组与假手术组相比,差异均有统计学意义(均P<0.05);实验组与模型组相比,差异均有统计学意义(均P<0.05)。结论β-Ecd介导β-catenin信号转导通路可延缓关节软骨退变,其作用机制可能是通过调控关节软骨组织的相关基因表达而实现的。Objective To explore the effects of β-ecdysterone(β-Ecd)on articular cartilage injury in mice with knee osteoarthritis mediated β-chain protein signaling transduction pathway.Methods Three month-old male C57 mice were randomly divided into three groups according by weight:model group,sham operation group and experimental group according by weight,10 mice in each group.Osteoarthritis mouse model were preparated of by operation.Two months after surgery,mice in experimental group were given subcutaneous injections of β-Ecd 5 mg·kg^-1;mice in model group and sham operation group were given normal saline,twice a day for 2 consecutive months.The gene expression of matrix metalloproteinase 13(MMP13),collagen X(ColX)and osteocalcin(OC)in articular cartilage tested by real time quantitative polymerase chain reaction.Results The relative expressions of MMP13 gene in sham operation group,model group and experimental group were 1.00±0.02,1.87±0.03 and 1.22±0.04;the relative expressions of of Col X gene in the three groups were 1.01±0.04,2.92±0.21,and 1.61±0.10,respectively;the relative expressions of OC gene in the three groups were 1.00±0.05,1.72±0.03,and 1.19±0.02,respectively.Comparison between sham operation group and model group,the difference of the factors were significantly(all P<0.05);comparison between experimental group and model group,the difference of the factors were significantly(all P<0.05).Conclusion β-Ecd mediated β-catenin signaling pathway delay the degeneration of articular cartilage,and the mechanism of action may be by regulating the expression of related genes in articular cartilage tissues.
