化妆品中防腐剂羟甲基甘氨酸钠的测定方法及甲醛释放机理研究

摘　　要：研究了化妆品中防腐剂羟甲基甘氨酸钠(Sodium Hydroxymethylglycinate, SHMG)在溶液中释放甲醛的机理并建立了测定方法。通过测定羟甲基甘氨酸钠在溶液中分解释放的甲醛量间接测定羟甲基甘氨酸钠的含量。羟甲基甘氨酸钠在溶液中快速分解释放甲醛,采用柱前衍生法使甲醛与2,4-二硝基苯肼反应后,经Inertsil ODS-SP液相色谱柱(250 mm×4.6 mm×5μm)分离测定,流动相为体积比60∶40的甲醇-水,检测波长为355 nm。结果表明,该测定方法检出限为10μg/g,羟甲基甘氨酸钠在1.102~110.250μg/m L范围内与相应的峰面积有良好的线性关系,加标回收率为98.0%~101.6%。羟甲基甘氨酸钠与甲醛的物质的量比为1︰1。该方法可用于测定化妆品中羟甲基甘氨酸钠的含量,填补了《化妆品安全技术规范》(2015年版)相关空白,有利于规范化妆品行业对甲醛释放体类防腐剂的使用。A method for determination of sodium hydroxymethylglycinate in cosmetics was established and its release mechanism was investigated. By determining the content of formaldehyde released from sodium hydroxymethylglycinate, the content of sodium hydroxymethylglycinate was determined indirectly. Sodium hydroxymethylglycinate degraded rapidly and released formaldehyde. After reacting with 2,4-dinitrophenylhydrazine and separated by Intersil ODS-SP column, formaldehyde was determined at 355 nm. The results showed that detection limit of this method was 10 μg/g, and the recovery rates varied from 98.0% to 101.6%. One molecular sodium hydroxymethylglycinate could release one molecular formaldehyde. The method which filled the blanks of Safety and Technical Standards for Cosmetics(2015 edition) can be used to determine the content of sodium hydroxymethylglycinate in cosmetics and to regulate the use of preservatives.
