摘  要:目的分析来自不同地区、具有不同遗传背景的献血浆者血浆抗体谱差异。方法收集血型、年龄、性别相匹配的来自巴马长寿地区有家族长寿背景和无家族长寿背景的血浆以及某东部地区的血浆各22份、即为3个大组,共66份血浆标本。每大组设置2个生物学重复,共6个小组,每个小组均为11份血浆各50μL等体积混合而成。以肠出血性大肠杆菌O157∶H7蛋白质组作为抗原库,利用蛋白质双向电泳技术和蛋白免疫印迹杂交技术获得不同血浆标本IgG的抗体谱,采用双向电泳图像分析软件进行差异分析。结果通过建立的方法获得了较高质量的血浆标本IgG抗体谱,3组标本中的IgG分别识别198、190和149个大肠杆菌蛋白质点;巴马地区有家族长寿背景和无家族长寿背景的血浆标本IgG抗体谱未见明显差异;但二者与东部地区血浆标本IgG抗体谱相比差异较为明显,如识别抗原数量不同,目标抗原点分布也不同。结论 3组血浆标本均含有针对大肠杆菌O157∶H7的广谱抗体;来自同一地区的血浆标本IgG抗体谱较为一致,来自不同地区人群的血浆标本IgG抗体谱差异明显,提示生活环境及遗传背景可能会影响人血浆中针对外来抗原的抗体的种类与多样性。Objective To analyze the differences of antibody repertoires against E. coli O157∶H7 in human plasma from different regions with different genetic backgrounds. Methods The plasma samples were collected from Bama and an eastern region in China(including 3 groups∶22 samples from longevity families in Bama, 22 samples from non-longevity families in Bama and 22 samples from the eastern region).Two biological duplicates were set up in each group, and a total of 6 sub-groups were set up.Each sub-group was composed of 11 plasma samples, each of which was mixed at the same volume of 50 μL.The antigenic library was composed of the proteins of E. coli O157∶H7.The two-dimensional electrophoresis and western blotting were used to study IgG antibody repertoires of the plasma samples, and the antibody repertoires were analyzed by a two-dimensional image analysis software.Results High-quality IgG antibody repertoires were obtained by the established method in our study.IgG from the samples of the 3 groups recognized 198, 190 and 149 E. coli protein spots respectively.There was no difference between the plasma samples from Bama′s longevity families and non-longevity families. However, when comparing the plasma samples between Bama and the eastern region, the differences were obvious, for example, the number of the antigens that IgG can recognize and the distribution of antigen points were different. Conclusion The plasma samples in all 3 groups contained broad-spectrum antibodies against E. coli O157∶H7;the IgG antibody repertoires were different between the plasma samples from different regions, which suggests that environment and genetic backgrounds might affect the species and diversity of human plasma antibodies against foreign antigens.
