摘  要:目的分析2017-2018年上海市长宁区流行性腮腺炎(流腮)病毒(Mumps virus,MuV)分离株的基因特征。方法对疑似流腮病例采集咽拭子标本,开展MuV核酸检测、SH和HN基因扩增和序列测定、基因型鉴定,分析分离株HN蛋白氨基酸序列变异。结果长宁区2017-2018年从123例疑似流腮病例中分离到5株MuV,获得4条SH基因序列和2条HN基因序列。5株MuV均鉴定为F基因型(中国优势流行株)。与中国疫苗株(S79株和JL株)相比,分离株HN蛋白第372和464氨基酸位点各增加了1个N-糖基化位点,第336、354-356、442、462、470、473-474和490位点存在氨基酸变异。结论2017-2018年长宁区MuV分离株属于中国优势流行的F基因型毒株,但存在一定的变异;建议持续加强流腮病毒学监测。Objective To analyze genetic characteristics of mumps virus(MuV)strains isolated in Changning district of Shanghai during 2017-2018.Methods We obtained throat swabs f rom suspected mumps cases to test for MuV nucleic acid and to amplif y and sequence the SH and HN target genes for determining genotypes.We analyzed variations of HN-gene-based amino acids.Results We isolated 5 MuV strains f rom 123 suspected mumps cases in Changning during 2017-2018 and obtained 4 SH sequences and 2 HN sequences.The 5 strains were identified as genotype F(Chinese predominant strain).One additional N-glycosylation site was seen at amino acid sites 372 and 464 of the HN genes in the isolates,with amino acid variations existing at sites 336,354-356,442,462,470,473-474,and 490,compared with the Chinese vaccine strains(S79 and JL strains).Conclusion The isolated MuV strains in Changning during 2017-2018 belonged to the Chinese predominant genotype F,with some variation.We should strengthen continuous MuV monitoring.
