摘  要:目的:探讨1.25(OH)2D 3对实验性自身免疫性神经炎(Experimental autoimmune neuritis,EAN)的治疗作用。方法:采用人工合成P0180-199肽段与完全弗氏佐剂混合免疫Lewis大鼠建立EAN模型,采用1.25(OH)2D 3灌胃处理,测大鼠体重,观察发病情况并进行行为学评分,HE染色观察坐骨神经炎细胞浸润情况,电镜检测坐骨神经脱髓鞘改变。RT-PCR和ELISA法检测大鼠白介素17(IL-17)、白介素10(IL-10)、转化生长因子-β(TGF-β)、干扰素(IFN-γ)RORrt、FoxP3的表达变化。结果:1.25(OH)2D 3干预可减轻EAN大鼠体重丢失情况、降低行为学评分,减少坐骨神经炎细胞浸润和减轻脱髓鞘,IL-17、IFN-γ、RORrt表达降低、IL-10、TGF-β和FoxP3表达增加。结论:1.25(OH)2D 3可改善EAN大鼠临床症状,机制可能与影响T细胞的数量和功能进而抑制促炎性因子表达有关,有望为格林巴利综合征的治疗提供新策略。Objective:To study the potential therapeutic effects of active Vitamin D 3(1.25(OH)2D 3)in the experimental autoimmune neuritis(EAN).Methods:EAN model was established by actively immunizing Lewis Rat with synthetic P0180-199 pepide and Freund's complete adjuvant.1.25(OH)2D 3 treatment was given,weight change of rats and clinical score were analyzed.HE staining was used to detect the inflammatory cell infiltration of sciatic nerve and demyelination of sciatic nerves was observed by Transmission electron microscope(TEM)at the same time.The expressions of inflammatory cytokines IL-17,IL-10,TGF-β,IFN-γwere detected by ELISA,and the expressions of Th17,Treg were examined by RT-PCR.Results:1.25(OH)2D 3 ameliorated body weight loss and myslin lesions.it decreased expression of inflammatory cytokines IL-17,IFN-γand RORrt while IL-10,TGF-βand FoxP3 were increased.Conclusions:1.25(OH)2D 3 can improve the clinical pathological changes of EAN rats,and the mechanism may be related to the changes of inflammatory cytokines.1.25(OH)2D 3 is expected to become a new strategy for the clinical treatment of GBS/EAN.
