摘 要:目的探讨miRNA-41调控c-myc在宫颈癌上皮间质转化(epithelial-mesenchymal transition,EMT)中的作用。方法HeLa宫颈癌细胞系随机分为3组,空白组、对照组与miRNA-41组,对照组与miRNA-41组分别转染miRNA-41 NC与miRNA-41 mimic,空白组不进行转染。qRT-PCR检测miRNA-41表达,MTT法检测细胞增殖指数,Transwell小室检测细胞侵袭指数,流式细胞术检测细胞周期,Western Blot检测c-myc与Vimentin表达。结果转染后24 h与36 h,miRNA-41组的miRNA-41表达水平显著高于空白组和对照组(P<0.05);细胞增殖指数与侵袭指数显著高于空白组和对照组(P<0.05);转染后36 h,miRNA-41组的G1期比例较对照组和空白组显著降低(P<0.05),S期+G2期比例显著增加(P<0.05)。转染后36 h,miRNA-41组的c-myc与Vimentin蛋白相对表达量显著高于对照组和空白组,空白组与对照组比较差异无统计学意义(P>0.05)。结论miRNA-41在宫颈癌细胞中的高表达能够促进c-myc与Vimentin蛋白表达,从而促使EMT的发生,调节细胞周期,提高细胞增殖性与侵袭性。Objective To investigate the role of miRNA-41 in the regulation of c-myc in epithelial-mesenchymal transition(EMT).Methods HeLa cervical cancer cell lines were randomly divided into three groups:blank group,control group and miRNA-41 group.The cells in control group and miRNA-41 group were transfected with miRNA-41 NC and miRNA-41 mimic,respectively,and the cells in blank group were not transfected.The expression levels of miRNA-41 were detected by qRT-PCR.The cell proliferation indexes were detected by MTT assay,and the cell invasion indexes were detected by Transwell chamber.Moreover the cell cycle was detected by flow cytometry,and the expression levels of c-myc and Vimentin were detected by Western Blot.Results At 24h and 36h after transfection,the expression levels of miRNA-41 in miRNA-41 group were significantly higher than those in blank group and those in control group(P<0.05).The cell proliferation indexes and invasion indexes in miRNA-41 group were significantly higher than those in blank group and those in control group(P<0.05).At 36h after transfection,the proportion of G1 in miRNA-41 group was significantly lower than that in control group and blank group(P<0.05),and the proportion of S+G2 phase was significantly increased(P<0.05).At 36h after transfection,the relative expression levels of c-myc and Vimentin in miRNA-41 group were significantly higher than those in control group and blank group,however,there were no significant differences between blank group and control group(P>0.05).Conclusion The high expression of miRNA-41 in cervical cancer cells can promote the expression of c-myc and Vimentin protein,so as to promote the occurrence of EMT,regulate cell cycle,and improve cell proliferation and invasiveness.
