摘 要:目的探讨A型肉毒毒素(BTXA)对增生性瘢痕组织成纤维细胞的作用以及酸性成纤维细胞生长因子(aFGF)和碱性成纤维细胞生长因子(bFGF)表达的影响。方法实验用的成纤维细胞来源于增生性瘢痕组织,在体外用含有0.2~1.6 U/ml浓度的BTXA的培养液孵育增生性瘢痕的成纤维细胞,采用MTT法测定细胞的存活率,罗丹明123染色在荧光显微镜下观察线粒体膜电位改变,H2DCFDA检测细胞内活性氧(reactive oxygen species,ROS)水平,评估其对瘢痕成纤维细胞的凋亡影响。利用RT-PCR分析用药前后aFGF和bFGF mRNA表达的变化。结果BTXA能够降低增生性瘢痕成纤维细胞的线粒体膜电位,改变线粒体膜通透性的稳定状态,并能够增加细胞内的活性氧(ROS)含量从而抑制增生性瘢痕成纤维细胞的增殖,同时诱导过度增殖的成纤维细胞凋亡;同时A型肉毒毒素也能够使两种成纤维细胞生长因子aFGF和bFGFmRNA的表达也呈下降趋势。结论BTXA对增生性瘢痕组织中成纤维细胞的增殖有明显抑制并诱导其凋亡,其作用机制可能与BTXA下调瘢痕组织中的成纤维细胞aFGF和bFGF两种因子的mRNA的表达有关。Objective To investigate the effects of botulinum toxin type A(BTXA)on fibroblasts in hypertrophic scar tissue and the expressions of acidic fibroblast growth factor(aFGF)and alkaline fibroblast growth factor(bFGF)in hyperplastic scar tissues.Methods The fibroblasts from in hyperplastic scar tissues were cultured by BTXA culture medium(0.2~1.6U/ml)in vitro.The?survival rate of hypertrophic scar fibroblasts was detected by MTT assay.And the changes of mitochondrial membrane potential were observed by rhodamine 123 staining under fluorescence microscope.Moreover,the level of reactive oxygen species(ROS)in cells was detected by H2DCFDA to evaluate its effect on apoptosis of fibroblasts.Furthermore,RT PCR was used to analyze the changes of the expressions of aFGF and bFGF before and after medication.Results BTXA could reduce the mitochondrial membrane potential of hypertrophic scar fibroblasts,change the stable state of mitochondrial membrane permeability,inhibit the proliferation of hypertrophic scar fibroblasts by increasing the content of intracellular reactive oxygen species(ROS),induce the apoptosis of hyperproliferating fibroblasts and decrease the expression ofaFGF and bFGF.Conclusion BTXA can significantly inhibit the proliferation of fibroblasts in hypertrophic scar tissue and induce their apoptosis.Its mechanism may be related to the expression of mRNA of aFGF and bFGF in down-regulated scar tissue.
