摘 要:微生物的生物量将直接影响脱胶酶的分泌量。以微生物的生物量作为指标,优化了两株剑麻脱胶酶的培养条件,探讨了营养条件(碳源、氮源)和环境条件(接种量、培养时间)对微生物生长的影响。最终优化的培养条件:TJM-3剑麻脱胶菌的最佳碳源是葡萄糖,最佳氮源为玉米浆,最优接种量7%,最佳菌种培养时间20 h;TJM-6剑麻脱胶菌的最佳碳源是乳糖,最佳氮源为牛肉膏,最优接种量5%,最佳菌种培养时间16 h。上述条件下培养菌种对应的生物量均提高较大,为下一步脱胶酶的分泌奠定良好的基础。Microbial biomass will directly affect the secretion of degumming enzyme.In this paper,microbial biomass was taken as an index to optimize the culture conditions of two strains of sisal degumming enzyme,and the effects of nutrient conditions(carbon source and nitrogen source)and environmental conditions(inoculation amount and culture time)on microbial growth were discussed.The optimal culture conditions were as follows:the optimal carbon source of TJM-3 sisal degummer was glucose,the optimal nitrogen source was corn pulp,the optimal inoculation amount was 7%,and the optimal culture time was 20 h.The best carbon source of TJM-6 sisal degum bacteria was lactose,the best nitrogen source was beef extract,the optimal inoculation amount was 5%,and the optimal culture time was 16 h.The biomass of the cultured strains increased significantly under the above conditions,which laid a good foundation for the secretion of degum enzyme in the next step.
