摘  要:目的:基于网络药理学方法,初步探讨益母草注射液止血调经的作用机制。方法:依托中药系统药理学数据库和分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)、STITCH数据库等,检索益母草注射液中水苏碱、胆碱和葫芦巴碱3种活性成分的作用靶点;通过药物治疗靶点数据库(therapeutic target database,TTD)和遗传药理学与药物基因组学数据库(pharmacogenetics and pharmacogenomics knowledge base,PharmGKB),检索与出血和月经不调等疾病相关的作用靶点,利用Cytoscape 3.6.1软件构建"化合物-化合物靶点"网络、"靶点-疾病"网络、"蛋白质-蛋白质相互作用"网络和"药物-化合物-靶点-通路"网络,借助DAVID v6.7平台进行基因本体(gene ontology,GO)功能富集分析和基于京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路富集分析,研究益母草注射液止血调经的作用机制。结果:益母草注射液的活性成分作用于"PTGS1""PTGS2""CYP2C9""CYP2C19"及"CYP2D6"等9个潜在靶点,这些靶点具有与血红素结合、与氧气结合、与铁离子结合和调节酶活性等分子功能,可用于产后出血、异常子宫出血及月经不调等妇科疾病的治疗;涉及KEGG通路5条,主要包括含血清素的神经突触通路、药物代谢-肝药酶CYP通路和亚油酸的新陈代谢通路等。结论:本研究初步预测了益母草注射液发挥止血调经作用的分子机制,为进一步揭示其作用机制提供了思路和线索。OBJECTIVE:To primarily explore the mechanism of motherwort injection for hemostasis and regulating menstruation based on network pharmacology.METHODS:Traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)and STITCH database were retrieved for the action targets of stachydrine,choline,trigonelline in motherwort injection.The therapeutic target database(TTD)and Genetic pharmacology and pharmacogenomics database(PharmGKB)were retrieved for the action targets of bleeding and irregular menstruation disease.Cytoscape 3.6.1 software was used to established the Compound-Compound Target network,Target-Disease network,Protein-Protein Interaction network and Drug-Compound-Target-Pathway network.Gene ontology(GO)functional enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis were carried out with the help of DAVID v6.7 platform to study the mechanism of motherwort injection for hemostasis and regulating menstruation.RESULTS:The active ingredients of motherwort injection could act on 9 potential targets,including PTGS1,PTGS2,CYP2 C9,CYP2 C19 and CYP2 D6.These targets had molecular functions such as binding with heme,binding with oxygen,binding with iron and regulating enzyme activity,and can be used in the treatment of postpartum hemorrhage,abnormal uterine bleeding,irregular menstruation and other gynecological diseases.There were 5 pathways,mainly included serotonin-containing synaptic pathway,drug metabolism-hepatic drug enzyme CYP pathway and linoleic acid metabolism pathway.CONCLUSIONS:This study preliminarily predict the molecular mechanism of motherwort injection for hemostasis and regulating menstruation,and provide the idea and clue for further revealing the mechanism of action on motherwort injection.
