摘 要:目的:基于网络药理学方法,初步探讨栀子柏皮汤治疗肝炎的作用机制。方法:通过建立栀子柏皮汤药物靶点数据集、肝炎相关疾病靶点数据集,构建栀子柏皮汤治疗肝炎的“成分-靶点-疾病”交互网络,并通过可视化分析、网络中各节点的拓扑参数分析,对各作用靶点进行京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路分析,预测栀子柏皮汤治疗肝炎的作用机制。结果:筛选出栀子柏皮汤中330个化学活性成分,4456个蛋白靶点,498个肝炎相关作用靶点。其中,栀子柏皮汤与肝炎相关疾病共同直接作用的蛋白靶点13个,共有24条相关通路,主要涉及肿瘤坏死因子信号通路、Jak-STAT信号通路及T细胞受体信号通路等。结论:栀子柏皮汤治疗肝炎的机制主要与其干预机体炎症反应和调节机体免疫有关。OBJECTIVE: To preliminarily probe into the mechanism of Zhizibopi decoction in the treatment of hepatitis based on network pharmacology. METHODS: Through the establishment of target data of Zhizibopi decoction and hepatitis,Ingredients-Target-Disease interactive network of Zhizibopi decoction in the treatment of hepatitis was developed. The mechanism of Zhizibopi decoction in the treatment of hepatitis was predicted by visualized analysis and topological parameter analysis,and the Kyoto encyclopedia of genes and genomes( KEGG) pathway was analyzed for each direct targets. RESULTS: Totally 330 active ingredients,4 456 directly active protein target points and 498 directly active protein target points in Zhizibopi decoction were screened out. Among them,totally 13 protein targets were directly affected by Zhizibopi decoction and hepatitis related diseases,with a total of 24 related pathways,mainly including tumor necrosis factor signaling pathway,signal transduction and transcription activator factor signaling pathway and T cell receptor signaling pathway. CONCLUSIONS: The mechanism of Zhizibopi decoction in the treatment of hepatitis is mainly related to the intervention of inflammatory reaction and the regulation of immunity.
