虎源猫杯状病毒的分离及其基因组遗传变异分析

摘　　要：从贵州省某动物园因发热、呼吸道等症状死亡的东北虎脾脏组织中分离得到1株猫杯状病毒(FCV),命名为FCVYH/16。采用RT-PCR的方法扩增了该病毒的全基因组,序列测定显示,FCV全基因组长7687 nt(GenBank序列登录号为KX815169),该基因组包含3个开放阅读框。基因组核苷酸序列分析显示,该毒株与12Q087-1、GD、SH和FB-NJ-13在一个大的分支上,而与疫苗株F4、F9和F65等不在一个分支上;其中与12Q087-1基因组同源性最高为80.5%,与疫苗株F9同源性最低为76.3%;衣壳蛋白氨基酸进化分析显示,与21株国内外代表株相比,该毒株的VP1氨基酸序列发生了9处新的单个氨基酸变异,5个区域的3个连续的氨基酸发生了变异,这些变异是否影响病毒毒力还需进一步研究。A feline calicivirus was isolated from the spleen tissue of Siberian tiger with fever and respiratory symptoms in Guizhou zoo,and was named as FCV-YH/16.The whole genome of FCV was amplified by RT-PCR.Sequence analysis showed that the length of FCV was 7687nt(GenBank number is KX815169).Genomic nucleotide sequence analysis showed that the FCV-YH/16 was in the large branch with 12Q087-1,GD,SH and FB-NJ-13,but not in a branch with vaccine strains F4,F9 and F65.The FCV-YH/16 shared the highest homology of 80.5%with 12Q087-1 strain,and the lowest homology of 76.3%with F9.Evolution analysis of capsid protein amino acid showed that the FCV-YH/16 strain had 9 new single amino acids,and three consecutive amino acid mutations occurred in five regions.Whether these mutations affect virulence of the virus needs further study.
