摘 要:为了研究鹿茸多肽(VAP)对人正常肝细胞系L-02肝损伤的保护作用。将人肝细胞系L-02暴露于酒精、四氯化碳、过氧化氢,MTT法检测细胞活力,Hoechst 33342和罗丹明123染色观察细胞核形态变化和线粒体膜电位变化,用2′7′-二氯二氢荧光素二乙酸(H2 DCFDA)染色观察细胞内活性氧(ROS)的产生。结果显示,在VAP存在的情况下,L-02暴露于酒精、四氯化碳、过氧化氢的细胞活力增加,VAP逆转了L-02细胞的核形态变化和线粒体膜电位的增加。表明VAP具有明显的抗肝损伤的作用,其机制可能与其抗氧化应激作用和降低ROS有关。To study the protective effect of Velvet Antler Polypeptide(VAP)against liver injury in human normal liver cell line L-02.The human liver cell line L-02 was exposed to alcohol、carbon tetrachloride,hydrogen peroxide and the cell viabilities were tested by MTT assay.Hoechst 33342 and rhodamine 123 staining were used to visulize nuclear morphological change and mitochondrial membrane potential alternation respectively.Intracellular reactive oxygen species(ROS)production was observed with 2′7′-Dichlorodihydrofluorescein diacetate(H2DCFDA)staining.Cell viability of L-02 exposed to alcohol、tetrachloride,hydrogen peroxide was increased in the presence of VAP,and VAP reversed the nuclear morphological change and an augmented mitochondrial membrane potential in L-02 cell.VAP protect against liver injury in vitro significantly.Its protective mechanism might be related to the anti-oxidative stress effect and decrease of the ROS.
