3种止痢型中兽药液体制剂中非法添加黏菌素的HPLC-PDA检查方法的建立

摘　　要：为建立止痢型中兽药液体制剂中非法添加黏菌素的高效液相二极管阵列色谱法,采用十八烷基键合硅胶为填充剂,流动相为乙腈∶硫酸钠磷酸水溶液(20∶80),硫酸钠磷酸水溶液∶硫酸钠水溶液(7 g/L)∶磷酸水溶液(6.8%)(50.5∶22.75)。检测波长为215 nm。结果显示,在该色谱条件下,3种止痢型中兽药液体制剂本底色谱峰对黏菌素E1的测定无干扰;黏菌素E1在0.04~2 mg/mL浓度范围内线性关系良好;杨树花口服液的检出限为10 mg/mL,板蓝根注射液及小檗碱注射液的检出限均为4 mg/mL;上述3种制剂的添加回收率在95.9%~97.9%之间,表明该方法回收率较好,准确度较高,可用于止痢型中兽药液体制剂中非法添加黏菌素的定性定量检测。The quantitative determination method of Colistin in the three veterinary Chinese medicine liquid preparation including Yangshuhua Koufuye,Banlangen Zhusheye,and Xiaobojian Zhusheye by HPLC-PDA was established.The mobile phase was acetonitrile:sodium sulfate phosphoric acid solution(20:80).The sodium sulfate phosphoric acid solution was sodium sulfate solution(7 g/L):phosphoric acid solution(6.8%)(50.5:22.75).The detection wavelength was 215 nm.Under this chromatographic condition,the detection of Colistin E1 was not disturbed by the chromatographic peaks from the three veterinary Chinese medicine liquid preparation.The linear relationship of Colistin E1 in 0.04~2 mg/mL concentration range was good.The detection limit of Yangshuhua Koufuye was 10mg/mL,and the detection limit of Banlangen Zhusheye and Xiaobojian Zhusheye was 4mg/mL.The recovery rate of the above three preparations ranged from 95.9%to 97.9%.The method has good recovery rate and high accuracy,which can be used for qualitative and quantitative detection of Colistin illegally added in veterinary Chinese medicine liquid preparation.
