摘 要:目的探究裂果薯皂苷单体Ⅰ通过转化生长因子-β1(transforming growth factor-β1,TGF-β1)对人肝癌细胞SMMC-7721侵袭和转移的影响。方法MTT法检测SMMC-7721细胞的增殖活性;用TGF-β1刺激细胞24 h,以TGF-β1/Smad信号通路抑制剂SIS3作阴性对照。细胞划痕实验和Tran-swell小室侵袭实验分别检测细胞迁移和侵袭能力;Western blot检测Smad7、Smad2/3、p-Smad2/3、E-cadherin、N-cadherin和Vimentin蛋白表达。结果SSPHⅠ能抑制SMMC-7721细胞的增殖,其24 h的IC50为1.72μmol·L^-1;与TGF-β1组相比,TGF-β1+SSPHⅠ组细胞迁移率和侵袭细胞数显著降低;Western blot实验结果显示,TGF-β1+SSPHⅠ组的Smad2/3,p-Smad2/3,N-cadherin和Vimentin蛋白的表达降低,Smad7和E-cadherin表达升高,结果与TGF-β1+SIS3组相同(P均<0.05)。结论SSPHⅠ能抑制SMMC-7721细胞侵袭和转移,其机制可能与抑制TGF-β1/Smad信号通路,阻断EMT发生有关。Aim To investigate the effect of saponin monomer I of Schizocapsa plantaginea Hance on inva-sion and metastasis of human hematoma cell line SMMC-7721 through TGF-β1.Methods The prolif-erating activity of SMMC-7721 cells was detected by MTT assay.Transforming growth factorβ1(TGF-β1)was used to stimulate cells for 24h,andSIS3,an inhibi-tor of TGF-β1/Smad signaling pathway,was used as negative control.The ability of cells on migration and invasion was examined by cell scratch assay and Tran-swell chamber invasion assay.The expressions of Smad7,Smad2/3,p-Smad2/3,E-cadherin,N-cadherin and Vimentin were detected by Western blot.Results SSPHⅠinhibited the proliferation of SMMC-7721 cells with IC50 of 1.72μmol·L^-1 in 24h.Compared with TGF-β1 group,the migration rate and the number of invasive cells in TGF-β1+SSPHⅠgroup were sig-nificantly reduced.The results of Western blot showed that the expression of p-Smad2/3,N-cadherin and Vi-mentin protein was down-regulated and the expression of Smad7 and E-cadherin up-regulated in TGF-β1+SSPHⅠgroup.These results were identical to TGF-β1+SIS3 group(P<0.05).Conclusions SSPHⅠin-hibits invasion and metastasis of SMMC-7721 cells.Its mechanism may be related to the inhibition ofTGF-β1/Smad signaling pathway,thereby blocking EMT.
