摘  要:目的:分析聚肌胞苷酸(Poly I:C)对氧糖剥夺损伤(oxygen-glucose deprivation,OGD)星形胶质细胞TLR3信号转导通路的影响.方法:对星形胶质细胞进行缺血缺糖培养12h建立氧糖剥夺损伤模型,观察组不同浓度Poly I:C预孵育对氧糖剥夺损伤星形胶质细胞Toll受体3(TLR3)、磷酸化干扰素调节因子(pIRF3)、磷酸化核因子(pNF-KB)蛋白表达的影响.结果:OGD组和Poly I:C+OGD组TLR3、pNF-KB表达水平均高于对照组(P﹤0.05),pIRF3与对照组对比无差异性(P﹥0.05).Poly I:C+OGD组与OGD组对比,pIRF3蛋白表达水平增加(P﹤0.05),pNF-KB表达水平降低(P﹤0.05),TLR3表达无差异性(P﹥0.05).结论:Poly I:C通过促进氧糖剥夺损伤星形胶质细胞pIRF蛋白的表达,进而上调下游抗炎因子干扰素-β的表到相关.Objective To analyze the effect of poly-inosinic acid(Poly I:C)on oxygen-glucose deprivation(OGD)astrocyte TLR3 signal transduction pathway.Methods The astrocyte was cultured for 12 hours after ischemia and glucose-deficient culture.The oxygen glucose deprivation injury model was established.The concentration of Poly I:C pre-incubation on oxygen glucose deprivation damage astrocyte Toll receptor 3(TLR3),phosphoric acid The effects of interferon regulatory factor(pIRF3)and phosphorylated nuclear factor(pNF-KB)protein expression.Results The expression levels of TLR3 and pNF-KB in OGD group and Poly I:C+OGD group were higher than those in control group(P<0.05).There was no difference between pIRF3 and control group(P>0.05).Poly I:Compared with OGD group,the expression of pIRF3 protein was increased(P<0.05),pNF-KB expression level was decreased(P<0.05),and TLR3 expression was not different(P>0.05).Conclusion Poly I:C can up-regulate the expression of pIRF protein in astrocytes by promoting oxygen glucose deprivation,and then up-regulate the expression of downstream anti-inflammatory factor interferon-β.
