摘 要:【目的】探讨葛根素预处理对脑缺血再灌注损伤(cerebral ischemia/reperfusion injury,CI/RI)沙鼠脑组织炎症反应的影响。【方法】将50只沙鼠随机分为葛根素3个剂量组(剂量分别为50、25、12.5 mg/kg)、模型组、假手术组,模型制备前3 d各组开始腹腔注射给药,采用双侧颈总动脉夹闭术制备CI/RI模型。采用卒中指数评估再灌注6 h内的神经症状,采用免疫组化法检测再灌注6 h后脑海马组织核因子-κB(nuclear factor-κB,NF-κB)、细胞间黏附分子1(intercellular cell adhesion molecule-1,ICAM-1)和基质金属蛋白酶9(matrix metalloproteinases-9,MMP-9)的表达。【结果】与模型组相比,葛根素高、中剂量预处理CI/RI沙鼠的卒中指数评分均有明显降低(P<0.01);与模型组相比,葛根素各剂量预处理均可明显下调脑海马组织NF-κB和ICAM-1的阳性表达,组间差异有统计学意义(P<0.05或P<0.01);葛根素高、中剂量脑海马组织MMP-9的表达有明显降低,与模型组相比差异有统计学意义(P<0.05)。【结论】葛根素预处理对沙鼠CI/RI具有脑保护作用,其作用机制可能是由于葛根素可抑制炎症因子表达,缓解脑组织的炎症反应。【Objective】To explore the effect of puerarin(PUE)pretreatment on inflammatory reaction of brain tissue in gerbils with cerebral ischemia-reperfusion injury(CI/RI).【Methods】A total of 50 gerbils were randomly divided into PUE high,medium and low dose groups(50,25 and 12.5 mg/kg,respectively),sham and model groups.PUE was intraperitoneally injected for 3 days before the model preparation.CI/RI model in gerbil was achieved by bilateral common carotid occlusion followed by 6-hour reperfusion.Stroke index was used to evaluate neurological symptoms of gerbils during 6-hour reperfusion.The activities of nuclear factor-κB(NF-κB),intercellular cell adhesion molecule-1(ICAM-1)and matrix metalloproteinases-9(MMP-9)in hippocampal tissue of CI/RI gerbils were examined by immunohistochemistry after 6-hour reperfusion.【Results】Compared with the model group,stroke index in PUE pretreatment groups(50 and 25 mg/kg)was markedly improved(P<0.01).Compared with the model group,the expression of NF-κB and ICAM-1 in hippocampi tissue in PUE pretreatment groups were significantly reduced(P<0.05 or P<0.01).PUE(50 and 25 mg/kg)markedly inhibited MMP-9 expression in hippocampal tissue compared with the model group(P<0.05).【Conclusion】PUE pretreatment can protect the brain tissue of CI/RI gerbils.Its action may be related with the inhibition of inflammatory factors and relieving inflammation in brain tissue.
