摘  要:在治疗肿瘤方面,纳米粒(Nanoparticles,NPs)制剂相比于传统的治疗药物有着绝对性的优势,但是由于肿瘤异常的微环境严重阻碍了NPs以有效的浓度到达肿瘤组织,导致治疗效果并不理想。在这一过程中,NPs首先需要通过血液循环系统到达肿瘤组织,然后跨越血管穿过间质才能靶向肿瘤细胞。该研究依次总结了NPs经历的每个阶段中所碰到的生理屏障,着重讨论了肿瘤异常的微环境和NPs的尺寸、形状、刚度以及表面修饰等性质对输运效率的影响,并且提出了克服这些屏障的方法,希望可以优化治疗方案,提高治疗疗效。In cancer treatment,nanoparticles( NPs) have absolute advantages over traditional drugs. However,the abnormal microenvironment of tumors seriously hinders NPs from reaching tumors at an effective concentration,resulting in unsatisfactory therapeutic effect. In this process,NPs first need to reach the tumor tissue through the blood circulation system,and then cross the blood vessels through the interstitium to target the tumor cells. This study summarized the physiological barriers encountered in each stage of NPs. The effects of abnormal microenvironment of tumors and the size,shape,stiffness and surface modification of NPs on transport efficiency are discussed emphatically. Meanwhile,methods to overcome these barriers are put forward. It is hoped that the therapeutic regimen can be optimized and improved.
