摘 要:肉碱脂酰转移酶1B(CPT1B)作为CPT1的一种重要同工酶--肌肉型肉碱脂酰转移酶1(MCPT1),在调节体内脂肪酸的氧化过程中起着重要作用,研究CPT1B基因的表达调控对于了解肉鸡脂肪沉积规律和脂质代谢调控机理具有重要意义。研究旨在阐明CPT1B基因在"花山麻鸡"组织表达和时序表达谱,为研究鸡体内脂肪代谢提供一定的基础资料。选取胚胎期到上市日龄的"花山麻鸡"为试验材料,在不同发育时间点采集胸肌、腿肌、心脏、肝脏组织样品,利用实时荧光定量方法,分析CPT1B基因在不同组织不同发育阶段表达规律。组织表达结果显示:在胚胎期(9日胚龄、12日胚龄、16日胚龄)和上市日龄(9周龄),CPT1B基因在肝脏的表达量显著高于在心脏、胸肌和腿肌的表达量(P<0.05);1日龄时,CPT1B在心脏的表达量显著提高(P<0.05),表达量与肝脏差异显著(P<0.05),且与胸肌和腿肌差异不显著;1周龄时,CPT1B在肝脏的表达量显著低于胸肌、腿肌和心脏(P<0.05);3周龄和4周龄时,CPT1B在肝脏、心脏、胸肌和腿肌的表达量差异不显著(P>0.05);5周龄和7周龄时,腿肌和胸肌的表达量显著低于出雏后的各个发育阶段(P<0.05)。时序表达谱结果显示,CPT1B基因在4种组织不同发育阶段变化趋势基本一致,但不同发育阶段表达量变化显著性不同。Carnitine fatty acyltransferase 1B(CPT1B), an important isoenzyme of CPT1, muscle carnitine fatty acyltransferase 1(MCPT1), plays an important role in regulating the oxidation of fatty acids in the body. Study on the expression regulation of CPT1B gene is of great significance for understanding the fat deposition rules and lipid metabolism regulation mechanism in broilers. The aim of this study was to elucidate the tissue expression and temporal expression profiles of CPT1B gene, and provide some basic data for studying the mechanism of fat metabolism in chicken. Huashan Partridge chickens from embryonic stage to market age were selected as experimental materials.Breast muscles, leg muscles, heart and liver tissue samples were collected at different development stage. The expression of CPT1B gene in different developmental stages of different tissues was analyzed by real-time fluorescence quantitative method. The results of tissue expression showed that the expression level of CPT1B gene in the liver was significantly higher than that in the heart, breast muscles and leg muscles during embryonic stage(9 days of embryo age, 12 days of embryo age, 16 days of embryo age) and market age(9 weeks of age)(P<0.05). At 1 day of age, the expression of CPT1B in the heart was significantly increased, and the expression level was significantly different from that in the liver(P<0.05), and was not significantly different from those of the breast and leg muscles. At 1 weeks of age, the expression of CPT1 B in the liver was significantly lower than that of the other three tissues(P<0.05). At 3 weeks of age and4 weeks of age, the expression levels of CPT1B in the liver, heart, chest muscles and leg muscles were not significantly different(P>0.05). At 5 weeks of age and 7 weeks of age, the expression of leg muscles and breast muscles was respectively significantly lower than that of other developmental stages after hatch(P<0.05).
