摘  要:目的研究内皮型一氧化氮合酶(e NOS)、促红细胞生成素(EPO)和肿瘤坏死因子-α(t TNF-α)基因多态性与青海地区藏族妊娠期高血压疾病发生的关系。方法选取2016年4月-2018年4月接受产前检查并分娩的60例藏族妊娠期高血压疾病患者为研究组,另选取同期分娩的60例正常孕妇为对照组。提取孕妇外周血液DNA,测量两组孕妇DNA浓度,使用琼脂糖凝胶电泳法观察DNA电泳图,合成TNF-α基因G308A、EPO基因T3541G和e NOS基因G894T上下游引物,使用琼脂糖凝胶DNA回收试剂盒对PCR产物进行切胶纯化,并进行酶切PCR产物、PCR纯化产物,对不同基因进行测序,并将结果输入美国国立生物技术信息中心数据库,判断PCR产物是否为目的基因片段,验证基因型分型结果。结果e NOS基因G894T多态性分布结果:研究组TT、GT型基因频率低于对照组,GG型基因频率高于对照组(P<0.05);研究组T等位基因频率低于对照组,G等位基因频率高于对照组(P<0.05);G等位基因为妊娠期高血压疾病患者易感基因。EPO基因T3541G多态性分布结果:研究组TG型基因频率高于对照组,TT型基因频率低于对照组(P<0.05);研究组T等位基因频率低于对照组,G等位基因频率高于对照组(P<0.05);G等位基因为妊娠期高血压疾病患者易感基因。TNF-α基因G308A多态性分布结果:研究组GG型基因频率低于对照组,GA型基因频率高于对照组(P<0.05);研究组G等位基因频率低于对照组,A等位基因频率高于对照组(P<0.05);A等位基因为妊娠期高血压疾病患者易感基因。结论TNF-α基因G308A、EPO基因T3541G、e NOS基因G894T多态性与青海地区藏族妊娠期高血压疾病的发生存在密切关系,EPO基因T3541G和e NOS基因G894T的G等位基因、TNF-α基因G308A的A等位基因为妊娠期高血压疾病患者易感基因。Objective To investigate the relationship between endogenous nitric oxide synthase(e NOS),erythropoietin(EPO)and tumor necrosis factor-α(TNF-α)gene polymorphisms and Qinghai tibetan gestational hypertensive disorder complicating pregnancy(HDCP)occurs.Methods Sixty Tibetan gestational HDCP patients who underwent antenatal examination and delivered in the hospital from April 2016 to April 2018 were selected as the study group.Sixty normal pregnant women who were delivered in the hospital were selected as the control group.DNA was extracted from peripheral blood of pregnant women,and measure the DNA concentration of two groups of pregnant women.DNA electrophoresis was observed by agarose gel electrophoresis,the results were input into the database of the National Biotechnology Information Center of the United States to determine whether the PCR product is the target gene fragment and verify the genotyping results.Results The results of G894 T polymorphism of e NOS gene were as follows:the genotype frequencies of TT and GT in the study group were lower than the control group,and the GG type gene frequency was higher than the control group(P<0.05).The T allele frequency of the study group was lower than control group,the G allele frequency was higher than control group(P<0.05),G allele was susceptibility gene for HDCP patients.The results of t3541 g polymorphism of EPO gene were as follows:the TG genotype frequency of the study group was higher than the control group,and the TT genotype frequency was lower than the control group,(P<0.05).The T allele frequency of the study group was lower than the control group,and the G allele frequency was higher than the control group(P<0.05).G allele was risk factors for HDCP patients.The results of G308 A polymorphism of TNF-αgene were as follows:the GG genotype frequency of the study group was lower than the control group,and the GA genotype frequency was higher than the control group(P<0.05).The G allele frequency of the study group was lower than the control group,and the a
