摘　　要：目的:观察间充质干细胞(MSC)治疗胶原诱导关节炎(CIA)大鼠的效果。方法:Ⅱ型胶原酶诱导关节炎大鼠,给予MSC尾静脉输注治疗,设MSC治疗组、甲氨蝶呤为阳性对照组、模型组与空白组,实验过程中每3天进行关节评分,实验结束后ELISA法测定各组大鼠血清中白介素-17(IL-17)、NF-κB配位体受体活化剂(RANKL)、可溶性蛋白骨保护素(OPG)和转化生长因子-β(TGF-β)水平,HE染色法观察关节破坏情况。结果:MSC与甲氨蝶呤作用相似,均能够有效缓解胶原诱导的关节肿胀;与模型组相比,MSC能够显著降低RANKL、TGF-β的水平,升高OPG的水平(P<0.05),而对IL-17影响不显著;组织学检验结果提示,MSC对CIA大鼠关节具有保护作用。结论:MSC能够提高通过抑制RANKL、TGF-β分泌,提高OPG水平,发挥对关节的保护作用,减轻CIA的关节肿胀程度,其治疗效果与甲氨蝶呤相似。Objects: To observe the therapeutic effect of umbilical cord mesenchymal stem cells in collagen induced arthritis(CIA) rats. Method: Type II collagenase induced rats were used. Mesenchymal stem cells(MSC) were injected through the tail vein of CIA rats, while others were treated with MTX or without any medication. Arthritis index was assessed through the whole procedure every three days. At the end of experiment, levels of IL-17, RANKL, OPG and TGF-β in all rat serum were determined through ELISA kits.The joint damages were observed by HE staining. Results: MSC could relieve the joint swelling caused by collagenase, which was similar to MTX. Compared with the model group, RANKL and TGF-β levels were suppressed by MSC, while OPG was significantly increased(P<0.05). MSC had limited function on secretion of IL-17. The histological results indicated that MSC had a protective effect on the joints of CIA rats.Conclusion: MSC could play an important role in protecting joint function and relieving joint swelling through increasing the level of OPG and inhibiting the secretion of RANKL and TGF-β, with potency equivalent to MTX.
