摘 要:利用3-氨丙基三乙氧基硅烷(ATPES)和N,N-二甲基十八烷基(3-三甲氧基硅丙基)氯化铵(DMOAP),对玻片基底表面进行烷基化修饰,再通过戊二醛(GA)作为桥联,将经过银纳米粒子(AgNPs)修饰的天蚕素B抗体(anti-CB)固定在玻片基底上,构建新型液晶生物传感器。当存在天蚕素B(CB)时,其可与anti-CB发生特异性免疫反应,从而固定在玻片基底表面。利用AgNPs具有较大的比表面积和良好的生物相容性,可以使得AgNPs周围的液晶分子垂直排列被极大地扰乱,从而起到局部光学信号放大的作用,达到检测更低浓度CB的目的。通过对CB浓度与其对应的图像灰度值之间的线性拟合发现,CB浓度在10~100 ng/mL之间时,CB浓度与其对应图像灰度值之间存在线性关系。实验结果表明,采用AgNPs对CB进行信号放大,其检出限可以低至1.02 ng/mL。The surface of the substrate was modified by alkyl of(3-aminopropyl)-triethoxysilane(APTES)and N,N-dimethyl-N-octadecyl-(3-aminopropyl)-trimethoxysilyl chloride(DMOAP).Then,the cecropin B antibody modified by silver nanoporticles(AgNPs)was immobilized on the substrate using glutaraldehyde(GA)as a bridge to construct a novel liquid crystal biosensor.When cecropin B is present,it will specifically immunoreact with cecropin B antibody and be fixed on the surface of the slide substrate.To make full use of AgNPs with large specific surface area and good biocompatibility,could make vertical alignment of liquid crystal molecules around AgNPs be greatly disturbed,thereby amplify local signal to achieve the purpose of detecting lower concentrations of cecropin B.By linear fitting between the cecropin B concentration and its corresponding image gray value,it was found that there was a linear relationship between the two things when the cecropin B concentration was between 10 and 100 ng/mL:y=0.353x+3.33.The experimental results showed that taking signal amplification of cecropin B by AgNPs,the detection limit was as low as 1.02 ng/mL.
