摘  要:为研究云南省境内特有的巨魾Bagarius yarrelli 6个野生群体的遗传多样性,在红河曼耗(Baya-M)、怒江坝(Baya-B)、怒江下游(Baya-N)、澜沧江上游(Baya-L)、景洪(Baya-J)、红河河口(Baya-H)6个点共采集72尾巨魾,每个点采集12尾,分析6个群体的ND4基因序列。结果表明:应用PCR技术扩增了巨魾线粒体DNA的ND4基因序列并测序,获得72条mtDNA DN4基因序列,巨魾ND4基因序列全长为1381 bp,共检测到501个核苷酸变异位点,核苷酸多样性为0.0019~0.0722;共有67种单倍型,单倍型间的平均相对遗传距离为0.098;将巨魾与三线纹胸鮡Glyptothorax trilineatus和长丝黑鮡Gagata dolichonema两种鱼类的ND4基因利用UPGMA中的Kimura 2-parameter法构建系统进化树,发现巨魾单聚为一支。研究表明,红河河口与曼耗群体之间的分化程度较小,亲缘关系较近,而澜沧江与景洪群体之间的遗传距离最大(0.156),说明澜沧江群体与景洪群体之间的分化程度较大,亲缘关系较远。A total of 72 mtDNA sequences of ND4 genes were analyzed in 12 samples of native fish Bagarius yarrelli collected from Manhao(Baya-M),the lower reaches of Nujiang River(Baya-N),Nujiangba-the upstream of Nujiang River(Baya-B),the upstream of Lanchangjiang(Baya-L),Jinhong-the lower reaches of Lanchangjiang(Baya-J),and Hekou-Honghe River(Baya-H).The results showed that the fish had ND4 gene with whole length of 1381 bp,501 nucleotide mutation loci in 1381 loci and 67 haplotypes in 72 sequences.The nucleotide diversity was found to be changed from 0.0019 to 0.0722 and the relative genetic distance of 0.098 among 67 haplotypes.UPGMA method with Kimura 2-parameter model in MEGA software was used to construct phylogenetic tree among ND4 genes studied in this study and other two fish,Glyptothorax trilineatus and Gagata dolichonema were applied as outgroup,which denoted that B.yarrelli gathered to a branch.The findings indicated that there was relative small degree of differentiation between Hekou-Honghe River(Baya-H)and Manhao(Baya-M),showing relatively close genetic relationship,and that relatively large genetic distance between Lancanjiang(Baya-L)and Jinghong(Baya-J)(0.156),showing relatively far genetic relationship.
