摘 要:王枣子为香茶菜属草本植物,具有较好的药用和食用价值.以当年王枣子种子为实验材料,研究快速打破种子休眠,提高发芽率的最佳方式.采收处于休眠期的王枣子种子,采用低温、赤霉素(GA3)、超声以及综合处理,获得破除王枣子种子休眠的条件.结果表明,低温、赤霉素(GA3)和超声处理均能一定程度解除王枣子种子休眠,但低温和GA3处理对破除种子休眠的效果有限,萌发率在2.5%~6.1%之间;而超声处理能有效促进种子萌发,处理5 min能将种子萌发率提高至30%以上.当基于单因素实验结果进行综合处理时,4℃低温处理14d、50 mg/L GA3浸泡6 h、超声处理5 min能将王枣子种子萌发率提高至37.6%,有效破除王枣子种子休眠.本研究获得休眠期王枣子种子萌发的优化条件,打破开展王枣子种子、萌发分析或组织培养受到的季节限制影响,有助于促进该药用植物的深入研究.Wangzaozi(Isodon amethystoides)is an Isodon medicinal herb with high medicinal and edible values.Currently related studies are mainly focused on identification of chemical compounds and pharmacological actions of this plant,less involved in the germination of dormancy seeds.Employing single factor experiments combined with comprehensive treatment,we optimized the physichemical parameters for the germination of Wangzaozi dormancy seeds.The results showed that coldstratification,gibberellin(GA3)and ultrasonic treatments promoted the germination of dormancy seeds to some extent.However,low temperature and GA3 treatment just had a limited effect on breaking the seed dormancy,having the germination rate between 2.5%~6.1%;by contrast,ultrasonic treatment effectively promoted the germination,with a germination rate up to30%after 5 min ultrasonic treatment.The comprehensive treatment based on single factor experiments enhanced the seed germination rate to 37.6%when 14 d coldstratification,6 h immersion with 50 mg/L GA3 and 5 min ultrasonic treatment was applied.Our study obtained the optimal conditions for the germination of dormancy Wangzaozi seeds,getting rid of the seasonal limitations of experimental materials for seed analysis,germination and tissue culture.
