摘  要:三叶青一般通过无性繁殖,极易感染病毒,造成其种性退化。为了探明三叶青携带病毒的种类,为其病毒病防控及病毒脱除提供科学依据,本研究在三叶青转录组和葡萄病毒序列的基础上,通过RT-PCR技术对怀玉山三叶青15种病毒(烟草花叶病毒,葡萄扇叶病毒,葡萄茎痘伴随病毒,葡萄卷叶病毒-1,葡萄卷叶病毒-2,葡萄卷叶病毒-3,葡萄卷叶病毒-4,葡萄卷叶病毒-5,葡萄卷叶病毒-6,葡萄卷叶病毒-7,葡萄卷叶病毒-9,葡萄病毒A,葡萄病毒B,葡萄病毒E,葡萄病毒F)进行RT-PCR验证。结果表明,怀玉山三叶青只检测出烟草花叶病毒,其他14种病毒没有检测到。本实验结果首次检测出了三叶青的主要病毒种类,可为三叶青脱毒苗的培育及其工厂化生产提供理论依据。Tetrastigma hemsleyanum Diels et Gilg is easy to infect virus and cause its species degeneration through asexual propagation.In order to find out the virus types of Tetrastigma hemsleyanum Diels et Gilg,and provide the scientific basis for its prevention and control of virus diseases and virus removal,on the basis of clover transcription group and grapevine virus sequence,15 viruses(Tobacco mosaic virus,Grapevine fanleaf virus,Grapevine rupestris stem pitting associated virus,Grapevine leafroll virus-1,Grapevine leafroll virus-2,Grapevine leafroll virus-3,Grapevine leafroll virus-4,Grapevine leafroll virus-5,Grapevine leafroll virus-6,Grapevine leafroll virus-7,Grapevine leafroll virus-9,Grapevine virus A,Grapevine virus B,Grapevine virus E and Grapevine virus F)of Huaiyushan Tetrastigma hemsleyanum Diels et Gilg were studied by RT-PCR in this paper.The results showed that only Tobacco mosaic virus was detected in Huaiyushan Tetrastigma hemsleyanum Diels et Gilg,but no other 14 viruses were detected.The main virus species of Huaiyushan Tetrastigma hemsleyanum Diels et Gilg were detected for the first time,which could lay a theoretical basis for the industrialized production of virus-free Tetrastigma hemsleyanum Diels et Gilg seedlings.
