摘 要:[目的]本文旨在探究亚硒酸钠(Na2SeO3)对大豆芽苗菜下胚轴中花青苷合成的作用及机制。[方法]以大豆芽苗菜(品种:‘东农690’)为试验材料,处理组为8μmol·L^-1 Na2SeO3根施培养的大豆芽苗菜,对照组为不加Na2SeO3的1/4 Hoagland培养液根施培养的大豆芽苗菜。采用酶标仪测定光照0、12、18、24和36 h时,大豆芽苗下胚轴花青苷总含量、H2O2、MDA、■含量和花青苷合成过程中关键酶(PAL、CHI、CHS、DFR、UFGT)活性,采用超高效液相色谱串联质谱法测定主要单体含量,利用实时荧光定量PCR技术(RT-qPCR)测定花青苷合成的结构基因(PAL、4CL、C4H、CHS、CHI、F3H、F3′H、DFR、ANS、UFGT)以及转录因子(MYB75)的相对表达水平。[结果]Na2SeO3处理组花青苷总含量和单体(除天竺葵素-3-O葡萄糖苷)含量显著高于对照组。H2O2、MDA、■含量随时间的变化趋势与花青苷总含量的变化趋势不一致。光照18 h时,对照组和Na2SeO3处理组中H2O2、MDA含量较高,分别是对照组的2.11和1.05倍。但花青苷总含量在光照12 h时已显著增加,约是对照组的1.72倍;光照18、24和36 h时,Na2SeO3处理组的H2O2、MDA、■含量显著高于对照组。Na2SeO3处理组花青苷合成的关键酶活性与关键基因的相对表达水平均显著高于对照组。[结论]与对照组相比,8μmol·L^-1 Na2SeO3处理组的大豆芽苗菜光照36 h,苯丙烷类代谢途径中花青苷合成的关键酶含量、关键结构基因和转录因子表达量均提高,下胚轴花青苷总含量提高了61.86%。[Objectives]This study aimed to investigate the effects and mechanisms of sodium selenite(Na2SeO3)on anthocyanin syn-thesis in the hypocotyl of soybean sprouts.[Methods]Glycine Max L.Merr.(the variety‘dongnong 690’)was applied as experimental material.The treatment group was soybean sprouts cultured with 8μmol·L^-1 Na2SeO3,and the control group was soybean sprouts cultured with 1/4 Hoagland without Na 2SeO 3.The content of total anthocyanin,H 2O 2,MDA,O·-2 and the activities of key enzymes(PAL,CHI,CHS,DFR,UFGT)were tested by enzyme-labeled instrument,the main monomers content were tested by high performance liquid chromatography tandem mass spectrometry(HPLC-MS),and the relative expressions of structural genes(PAL,4CL,C4H,CHS,CHI,F3H,F3′H,DFR,ANS,UFGT)and transcription factor(MYB 75)were tested by RT-PCR.[Results]Compared with the control group,Na2SeO3significantly increased the total amount and monomers(except Pelargonidin-3-O-glucoside)content of anthocyanin in the hypocotyl of soybean sprouts.The variation trend of H 2O 2,MDA,O·-2 content was inconsistent with that of anthocyanin total content.At the illumination time of 18 h,the contents of H 2O 2 and MDA in the control group and the Na 2SeO 3 treatment group were higher,which were 2.11 times and 1.05 times those of the control group,respectively.However,the total anthocyanin content increased significantly at the illumination time of 12 h,about 1.72 times that of the control group.At the illumination time of 18,24,36 h,the contents of H 2O 2,MDA and O·-2in Na2SeO3 treatment group were significantly higher than those in the control group.The key enzyme activity and relative expression level of key genes in anthocyanin synthesis in Na2SeO3 treatment group were significantly higher than those in the control group.[Conclusions]Compared with the control group,at the illumination time of 36 h,the 8μmol·L^-1 Na2SeO3 treatment promoted the expression of key enzymes,key structural genes and transcription factors in anthocyanin synthesis in phenylpropane
