摘 要:介电电泳(DEP)是一种基于尺寸或电性质分离和鉴定悬浮在介质溶液中的微颗粒技术。为了克服传统接触式介电泳电极污染样品和芯片工艺复杂等问题,采用液体电极代替金属电极或薄膜电极。在该芯片中,电极通过电介质阻挡层电容耦合到流体通道,用非接触式液体沟道电极代替传统接触式金属电极。实验中,分别考察了电压和频率对液体电极捕获微球和细胞的影响。结果表明,电压和频率对液体电极捕获微球和细胞均有重大影响,同时没有发现被捕获的细胞有裂解现象,活力良好。芯片中不同细胞的电旋转速度明显不同,说明该芯片具有通过微颗粒/细胞自身独特电特性捕获和识别不同微颗粒/细胞的能力。Dielectrophoresis(DEP)is a technique for separating and identifying micro-particles and cells suspended in the medium solution based on size or electrical properties.In order to solve the problems that the electrode of traditional contacting-type dielectrophoresis can contaminate sample and the chip production process is complicated,liquid electrode is used instead of metal electrode or film electrode.In the chip,the electrode is coupled to the fluid channel through the capacitance of dielectric barrier layer,and the non-contact liquid channel electrode replaces traditional contact metal electrode.In the experiments,the effects of voltage and frequency on the capture of microspheres and cells are investigated.The results show that both voltage and frequency have significant effects on the capture of microspheres and cells by the liquid electrode,and no cracking phenomenon is found in the captured cells.The electrical rotation speed of different cells in the chip is obviously different,indicating that the chip has the ability to capture and recognize different micro-particles/cells through their own unique electrical characteristics.
