摘 要:研究通过GC-MS法分析了大马士革玫瑰(Rosa damascena)精油和苦水玫瑰(Rosa sertata×Rosa rugosa)精油的化学成分。将ICR小鼠分成对照组、地西泮组和精油组,采用高架十字迷宫模型(elevated plus-maze,EPM)评价了急性嗅吸不同浓度2种精油对小鼠抗焦虑的影响。通过高架十字迷宫和明暗箱(light-dark box,LDB)模型评价了长期嗅吸苦水玫瑰精油对抗焦虑的影响。精油成分分析显示,苦水玫瑰精油的主要成分中香茅醇(37.36%)和香叶醇(12.06%)含量均高于大马士革玫瑰精油中香茅醇(16.87%)和香叶醇(11.63%)含量。急性嗅吸实验结果显示,与对照组(开臂时间%:1.06±0.70,开臂次数%:0.68±0.49)相比,低浓度(10μL/mL)苦水玫瑰精油(开臂时间%:14.05±6.39,开臂次数%:16.16±6.19)具有显著缓解小鼠焦虑的作用(P<0.05),而大马士革玫瑰精油则不具有抗焦虑功效。长期嗅吸实验结果显示,与对照组相比,苦水玫瑰精油可以显著缓解小鼠在EPM(开臂时间%:14.05±6.39,开臂次数%:17.52±2.60)和LDB(精油组:明箱时间%:48.04±2.28,明箱距离%:46.26±3.19)模型中的焦虑行为(P<0.05)。In this study,GC-MS was used to analyze the chemical constituents of essential oils of the Damask rose(Rosa damascena)and Kushui rose(Rosa sertata×Rosa rugosa).ICR mice were divided into control group,diazepam group and essential oil group.Elevated plus-maze(EPM)test was used to evaluate the acute anxiolytic effect of the two kinds of rose oils in different concentrations.EPM and light-dark box(LDB)tests were used to evaluate the chronic anxiolytic effect of Kushui rose oil.The GC-MS results showed that the content of citronellol(37.36%)in Kushui rose oil was higher than that of Damask rose oil(16.87%).The content of geraniol(12.06%)in Kushui rose oil was higher than that of Damask rose oil(11.63%).The behavioral tests results showed that low concentration(10μL/mL)of Kushui rose oil(open arm time%:14.05±6.39,open arm entry%:16.16±6.19)showed significant anxiolytic effect compared to the control(open arm time%:1.06±0.70,open arm entry%:0.68±0.49)(P<0.05).All of the three concentrations of Damask rose oil showed no significant efficacy.Chronic exposure of Kushui rose oil significantly reduced the anxiety behaviors of the mice in the EPM test(open arm time%:14.05±6.39,open arm entry%:17.52±2.60)and LBD test(light box time%:48.04±2.28,open box distance%:46.26±3.19)(P<0.05).
