摘 要:目的探讨生命早期应用抗生素对小鼠体重、脂代谢以及胰岛素的影响。方法将2周龄昆明小鼠144只随机分为静脉给药对照组(注射生理盐水)、静脉单独给药组(头孢噻肟)、静脉联合给药组(头孢噻肟联合氨苄西林舒巴坦)、口服对照组(生理盐水)、口服单独给药组(单用头孢噻肟)、口服联合给药组(头孢噻肟联合氨苄西林舒巴坦),每组24只,分别予相应干预1周,后饲养3个月。分别于1月末、2月末、3月末每组剖杀8只小鼠,分析各组小鼠体重、脂代谢、脂多糖及胰岛素的情况。结果与静脉对照组小鼠相比,静脉单用组和静脉联用组小鼠体重在第1个月末、第2个月末有明显增加,差异有统计学意义(P<0.05),小鼠胰岛素水平大致呈先降低再升高趋势(P<0.05),脂代谢表现为高密度脂蛋白(HDL)呈先降低再升高趋势(P<0.05),低密度脂蛋白(LDL)、甘油三酯(TG)大致呈先升高再降低趋势(P<0.05)。静脉给药方式的3组小鼠总胆固醇比较,差异无统计学意义(P>0.05)。小鼠体质量、胰岛素水平、脂代谢水平在口服抗生素两组小鼠与其对照组比较,差异无统计学意义(P>0.05)。结论生命早期静脉应用抗生素会在较长时间内促进小鼠体重增加,LDL、TG水平升高,HDL水平下降,下调胰岛素水平,这种影响在口服给药中不明显。Objective To investigate the effect of the antibiotic application in early life on body mass,lipid metabolism and insulin in mice.Methods A total of 1442-week-old Kunming mice were randomly divided into six groups including the intravenous administration control group with normal saline,intravenous administration group with cefotaxime,combined intravenous administration group with cefotaxime and ampicillin sulbactam,oral administration control group with normal saline,oral administration group with cefotaxime and combined oral administration group with cefotaxime and ampicillin sulbactam,and each group included 24 mice in each group.All the mice in each group were given the intervention for 1 week and then fed for 3 months.Every 8 mice in each group were sacrificed at the end of the 1 st,2 nd and 3 rd month respectively and their body weight,lipid metabolism,lipopolysaccharide and insulin were analyzed.Results Compared with the intravenous administration control group,the body mass was significantly increased(P<0.05)and the insulin level decreased at first and then increased at the end of the 1 st and 2 nd month in the intravenous administration group and the combined intravenous administration group(P<0.05).The lipid metabolism showed that high density lipoprotein(HDL)decreased at first and then increased(P<0.05)and low density lipoprotein(LDL)and triglyceride(TG)increased at first and then decreased(P<0.05).There were no significant differences in total cholesterol(TC)between the three groups of intravenous administration(P>0.05),and there were no significant differences in body weight,insulin level and lipid metabolism between the three groups of oral administration(P>0.05).Conclusion The intravenous administration with antibiotics in early life can increase the body mass and the levels of LDL and TG,decrease the HDL level and down-regulate the insulin level over a long period of time in mice,and the effect was not significant in the oral administration.
