摘  要:糖基化是蛋白质翻译后修饰的关键过程,寻找该过程中的特征性改变可以为疾病的早期诊断、治疗、预后评估提供关键信息。异常的糖基化修饰与肾脏疾病的发生、发展密切相关,例如IgA肾病、糖尿病肾病和常染色体显性多囊肾病等。近来,质谱糖蛋白分离、富集技术的进展大大促进了糖蛋白组学检测的精确性,这为临床进一步认识肾脏疾病的发病机理提供新的技术平台。由于糖蛋白的低丰度和糖链的不均一性,在糖蛋白组检测过程中仍有许多亟待解决的问题,故本文就常用的糖蛋白组学分离富集技术及其在肾脏疾病研究中的进展进行综述。Glycosylation is a key process for post-translational modification of proteins.By looking for its characteristic changes,we can provide significant information for early diagnosis,treatment and prognosis evaluation.Abnormal glycosylation modification is closely related to the occurrence and development of renal diseases,including IgA nephropathy,diabetic nephropathy and autosomal dominant polycystic kidney disease.In recent years,the advancement of mass spectrometry glycoprotein separation and enrichment technology has greatly promoted the accuracy of glycoproteomics detection,which provides a new technology platform for further understanding the pathogenesis of kidney disease in clinical work.However,considering the low abundance of glycoprotein and the heterogeneity of glycan chain,there are still many obstacles in the glycoproteomics detection process.This article reviews the four commonly used glycoproteomic techniques and their pros and cons,along with their application in the study of renal diseases.
