摘 要:目的探讨雷公藤甲素对尤因肉瘤(ES)细胞增殖、迁移和凋亡的影响。方法将处于对数生长期的ES细胞分为5组:空白组(等量培养液)、对照组(阿霉素稀释液6.5μmol·L^-1)和低、中、高3个剂量实验组(雷公藤甲素稀释液10,30,50 nmol·L^-1)。以CCK8检测各组细胞增殖的情况;以Transwell检测各组细胞迁移情况;以流式细胞术检测各组细胞凋亡情况;以蛋白质印迹法检测各组p-蛋白激酶B(AKT)、B淋巴细胞瘤-2(Bcl-2)和Bcl-2相关蛋白(Bax)的蛋白相对表达(灰度比值)情况。结果空白组、对照组和中、高2个剂量实验组的细胞增殖率分别为(0.94±0.14)%,(0.55±0.21)%,(0.48±0.18)%和(0.22±0.11)%;这4组的细胞迁移数分别为396.21±28.45,165.64±19.21,182.24±33.41和195.54±24.35;这4组的p-AKT相对表达量分别为1.14±0.14,0.55±0.23,0.64±0.27和0.72±0.35;这4组的Bcl-2相对表达量分别为0.89±0.32,0.29±0.17,0.31±0.33和0.34±0.19;这4组的Bax相对表达量为0.34±0.21,0.73±0.15,0.67±0.18和0.74±0.26。上述指标:对照组和中、高2个剂量实验组与空白组相比,差异均有统计学意义(均P<0.05);对照组与低剂量实验组(除增殖率外)相比,差异均有统计学意义(均P<0.05)。结论雷公藤甲素可以抑制ES细胞的增殖和迁移,并能促进ES细胞凋亡,其机制可能是通过AKT信号通路实现的。Objective To investigate the effects of triptolide on proliferation,migration and apoptosis of Ewing sarcoma(ES)cells.Methods The ES cells in logarithmic growth stage were divided into five groups:blank group(equal volume culture medium),control group(adriamycin diluent 6.5μmol·L^-1)and low,medium,high doses of the experimental groups(triptolide diluent 10,30,50 nmol·L^-1).The proliferation of each group was detected by CCK8.The migration of each group was detected by Transwell.The apoptosis of each group was detected by flow cytometry.The relative expressions of p-protein kinase B(p-AKT)and B lymphocyte tumors-2(Bcl-2),Bcl-2 related protein(Bax)proteins were detected by Western blot.Results The cell proliferation rates in blank group,control group,experimental-M,-H groups were(0.94±0.14)%,(0.55±0.21)%,(0.48±0.18)%,(0.22±0.11)%,respectively;the number of cell migration in the four groups were 396.21±28.45,165.64±19.21,182.24±33.41,195.54±24.35,respectively;the relative expression levels of p-AKT in the four groups were 1.14±0.14,0.55±0.23,0.64±0.27,0.72±0.35,respectively;the relative expression of Bcl-2 in the four groups were0.89±0.32,0.29±0.17,0.31±0.33,0.34±0.19,respectively;the relative expression of Bax in the four groups were 0.34±0.21,0.73±0.15,0.67±0.18,0.74±0.26,respectively.Comparison between control group,experimental-M,-H groups and blank group,the difference of the factors were statistically significant(all P<0.05).Comparison between control group and experimental-L group(except for proliferation rates),the difference of the factors were statistically significant(all P<0.05).Conclusion Triptolide can inhibit the proliferation and migration of ES cells and promote the apoptosis of ES cells,possibly through the AKT signaling pathway.
