摘  要:目的研究七氟醚预处理对脓毒症模型小鼠NALP3炎性体的影响。方法C57BL/6N健康雄性小鼠84只,采用随机数字表法,分为七氟醚对照组、七氟醚脓毒症组、氧气对照组、氧气脓毒症组,每组21只,参照文献,采用盲肠结扎穿刺法制备小鼠脓毒症模型。观察比较四组小鼠生存率;肝、肺以及肾脏组织的炎性评分;RT-PCR方法检测血液、腹腔灌洗液、肺灌洗液NALP3、Caspase1及ASC mRNA表达。结果七氟醚脓毒症组小鼠生存率低于七氟醚对照组(66.67%比100%,P<0.05),氧气脓毒症组小鼠生存率低于氧气对照组(55.56%比100%,P<0.05)。与七氟醚对照组比较,七氟醚脓毒症组小鼠肺、肝、肾组织炎症评分明显升高[(2.08±0.21)分比(0.83±0.44)分、(2.33±0.57)分比(0.87±0.46)分、(2.25±0.49)分比(0.71±0.24)分,P均<0.05];与氧气对照组比较,氧气脓毒症组小鼠肺、肝、肾组织炎症评分明显升高[(3.58±0.32)分比(0.82±0.37)分、(3.24±0.46)分比(0.92±0.33)分、(3.06±0.47)分比(0.67±0.22),P均<0.05];七氟醚脓毒症组小鼠肺、肝、肾组织炎症评分明显低于氧气脓毒症组[(2.08±0.21)分比(3.58±0.32)分,(2.33±0.57)分比(3.24±0.46)分,(2.25±0.49)分比(3.06±0.47)分,P均<0.05]。脓毒症组小鼠血NALP3、Caspase-1、ASC mRNA表达量显著高于对照组[NALP3:(1.58±0.42)比(1.10±0.49)、(1.47±0.54)比(1.06±0.36);Caspase-1:(1.93±0.42)比(1.06±0.44)、(1.76±0.32)比(1.03±0.29);ASC mRNA:(1.92±0.47)比(1.06±0.46)、(1.65±0.35)比(1.03±0.29),P均<0.05];与氧气脓毒症组比较,七氟醚脓毒症组小鼠血NALP3、Caspase-1、ASC mRNA表达显著下调[NALP3:(1.47±0.54)比(1.58±0.42);Caspase-1:(1.76±0.32)比(1.93±0.42);ASC mRNA:(1.65±0.35)比(1.92±0.47),P均<0.05]。氧气脓毒症组小鼠腹腔灌洗液NALP3、Caspase-1、ASC mRNA表达量显著高于氧气对照组[NALP3:(2.34±0.25)比(1.73±0.62)、(2.07±0.33)比(1.91±0.40);Caspase-1:(2.31±0.26)比(1.77±0.72)、(2.19±0.24)比(1.95±0.71);ASC mRNA:(2Objective To explore the Effects of sevoflurane pretreatment on NALP3 inflammasome in septic mice. Methods Eighty-four C57 BL/6 N healthy male mice were randomly divided into sevoflurane control group,sevoflurane sepsis group, oxygen control group and oxygen sepsis group, 21 animals in each group. The sepsis model of mice was established by cecum ligation and puncture method, according to the literatures. The survival rate, inflammatory scores of liver, lung and kidney were observed and compared. mRNA expression of NALP3, caspase 1 and ASC in blood, peritoneal lavage fluid and lung lavage fluid were detected by RT-PCR. Results The survival rate of sevoflurane sepsis group was lower than that of sevoflurane control group(66.67% vs 100%, P <0.05), and that of oxygen sepsis group was lower than that of oxygen control group(55.56% vs 100%, P<0.05).Compared to the sevoflurane control group, the inflammatory scores of lung, liver and kidney tissues in sevoflurane sepsis group increased significantly, i.e., lung: [(2.08±0.21) vs(2.08±0.21)], liver: [(2.33±0.57) vs(0.87±0.46)],kidney: [(2.25±0.49) vs(0.71±0.24)], P<0.05, respectively. Compared to the oxygen control group, the inflammation scores of lung, liver and kidney tissues in the oxygen sepsis group were significantly increased, i.e., lung:[(3.58±0.32) vs(0.82±0.37)], liver: [(3.24±0.46) vs(0.92±0.33)], kidney: [(3.06±0.47) vs(0.67±0.22)], P<0.05, respectively. And those of in sevoflurane sepsis group were significantly lower than those in oxygen sepsis group, i.e.,lung: [(2.08±0.21) vs(3.58±0.32)], liver: [(2.33±0.57) vs(3.24±0.46)], kidney: [(2.25±0.49) vs(3.06±0.47)], P<0.05, respectively. For mRNA expressions of NALP3, caspase 1 and ASC in the blood, the sepsis group were significantly higher than those of the control group, i.e., NALP3: [(1.58±0.42) vs(1.10±0.49),(1.47±0.54) vs(1.06±0.36)], Caspase-1: [(1.93±0.42) vs(1.06±0.44);(1.76±0.32) vs(1.03±0.29)], ASC: [(1.92±0.47) vs(1.86±0.52),(1.65±0.35) vs(1.03±0.29)], P<0.05, respecti
