摘  要:目的:建立反相高效液相-二极管阵列检测法(HPLC-DAD),采用多波长切换法同时测定倍生颗粒中二苯乙烯苷、阿魏酸、人参皂苷Rg1、人参皂苷Re、人参皂苷Rb1、丹参酮ⅡA的含量测定方法。方法:采用Waters XBridge-C18色谱柱(4.6 mm×150 mm,5μm),流动相:乙腈-0.1%磷酸水,梯度洗脱,流速:1.0 m L/min,检测波长:320 nm(0~25 min)、203 nm(25~75 min)、270 nm(95~110 min),柱温:30℃。结果:二苯乙烯苷、阿魏酸、人参皂苷Rg1、人参皂苷Re、人参皂苷Rb1、丹参酮ⅡA分别在0.101~0.804、0.178~1.421、0.225~1.800、0.557~4.456、0.390~3.120、0.100~0.796μg范围内呈良好的线性关系,r分别为0.999,3、0.999,3、0.999,0、0.998,6、0.999,2、0.999,9;平均回收率分别为98.11%、97.03%、98.86%、99.11%、101.10%、99.81%。结论:本方法操作简便、灵敏度高、准确性及重复性好,可作为倍生颗粒的质量控制方法。Objective: To establish a reversed-phase high-performance liquid-diode array detection(HPLCDAD) method for simultaneous determination of stilbene glycosides, ferulic acid, ginsenoside Rg1, ginsenoside Re,ginsenoside Rb1 and tanshinone IIA in Beisheng granules by multi-wavelength switching method. Methods: The waters XBridge-C18 column(4.6 mm×150 mm, 5 μm) was used. The mobile phase was acetonitrile-0.1% phosphoric acid water, gradient elution, with the flow rate was 1.0 mL/min. The detection wavelengths were 320 nm(0-25 min),203 nm(25-75 min), 270 nm(95-115 min),and the column temperature was 30 ℃. Results: There was a good linear relationship among stilbene glycoside, ferulic acid, ginsenoside Rg1, ginsenoside Re, ginsenoside Rb1,tanshinone ⅡA in the range of 0.101-0.804, 0.178-1.421, 0.225-1.800, 0.557-4.456, 0.390-3.120, 0.100-0.796μg, r were 0.999,3, 0.999,0, 0.998,6, 0.999,2, 0.999,9, respectively. The average recoveries were 98.11%,97.03%, 98.86%, 99.11%, 101.10% and 99.81% respectively. Conclusion: The method is simple, sensitive, accurate and reproducible, and can be used for the quality control of Beisheng granules.
