摘  要:为评价伪狂犬病病毒(PRV)变异株和经典株对小鼠的致病力差异,本研究将PRV变异株(PRVTJ)和经典株(PRVSC)以103 TCID50的剂量经肌肉注射途径接种小鼠之后,通过观察小鼠的临床症状、记录小鼠的体质量变化和死亡率,利用荧光定量PCR(qPCR)检测小鼠各组织中病毒基因拷贝数,分析小鼠各组织脏器的病理学变化,系统评价PRV变异株和经典株的致病力差异。研究结果显示,接种PRVTJ和PRVSC的小鼠在临床表现上无明显差异,但接种PRVTJ的小鼠体质量下降明显,死亡率相对更高;qPCR检测结果显示,PRVTJ对小鼠的组织嗜性增强,尤其是对脑组织的嗜性增强;病理组织学分析结果显示,接种PRVTJ的小鼠脑组织的炎性病变更明显。本研究初步证实,PRVTJ对小鼠的致病力增强,且其毒力增强的主要原因是对小鼠的神经嗜性增强。本研究为PRV变异株致病力增强机制研究奠定了基础。To assess the pathogenicity of pseudorabies virus variant and classical strains,mice were infected intramuscularly with 103 TCID50 PRV variant strain PRVTJ or PRV classical strai PRVSC strain.The pathogenicity of different PRV strains was evaluated based on clinical signs,weight changes,survival rate,viral distribution and histopathological changes in diverse tissues of mice.The results demonstrated that there was no significant difference in the clinical signs of mice infected with either PRVTJ and or PRVSC,but the mice infected with PRVTJ had remarkable decline of weight and higher mortality.The results of qPCR revealed that PRVTJ exhibited enhanced tissue tropism to mouse tissues,especially brain tissues.Histopathological analysis showed that inflammatory lesions in the brain tissues of PRVTJ-infected mice were more significant.In general,this study demonstrated that the main reason for the enhanced virulence of PRVTJ is due to the increased neurotropism to mice.These results lay the foundation for further study on the detailed molecular mechanism of the enhanced pathogenicity of PRV variants.
