摘 要:采用沉淀法制备紫杉醇白蛋白微粒,采用高效液相色谱法(HPLC)测定紫杉醇质量浓度,以二氯甲烷为有机相,采用萃取法分离白蛋白微粒与游离药物,测定其包封率。结果表明:紫杉醇在10~100 mg·L-1范围内线性良好,平均回收率可达到97.6%~101.0%;有机溶剂萃取法测定紫杉醇白蛋白微粒包封率的平均回收率,达94.80%~101.67%。这种方法测定紫杉醇白蛋白微粒回收率准确可靠,适用于沉淀法制备的白蛋白微粒的包封率测定。In order to establish a method to determine the encapsulation efficiency of paclitaxel-albumin(PTX-BSA),microparticles was produced by precipitation method.Paclitaxel(PTX)content(mass concentration)was determined by HPLC.Separating free drugs from microparticles system was achieved by organic solvent extraction method,and methylene dichloride as the extracting solvent.PTX is well separated by HPLC.There is a quite good linear relationship between concentration and peak area in the range of 10-100μg·mL-1,the recovery rate is 97.6%-101.0%.The organic solvent extraction method can effectively separate the free drugs from the microparticles suspension,the sample recovery rate is 94.80%-101.67%.The organic solvent extraction method can be accurate and effective to determine the PTX-BSA microparticles entrapment efficiency.
