摘 要:为探究营养性肥胖对小鼠生殖器官发育及孕酮膜受体基因在子宫、卵巢、输卵管中表达的影响,试验通过饲养昆明雌鼠40只,随机分为高脂组(n=20)和普通组(n=20),分别给予高脂饲料与普通饲料,构建小鼠的营养性肥胖模型。小鼠喂养8周后,通过测量体重,高脂组比对照组提高28.47%(P<0.01),肥胖模型构建成功,并采集子宫、卵巢、输卵管进行称重,再通过定量PCR测定mPRα、mPRβ、mPRγ在子宫、卵巢、输卵管的表达。结果发现:8周后,高脂小鼠子宫和卵巢体重较对照组分别降低68.39%和75.27%(P<0.05);高脂组子宫和输卵管中mPRα表达量较对照组分别降低8.65%和46.50%(P<0.05),高脂组卵巢中mPRα表达量较对照组降低45.25%(P>0.05);高脂组小鼠卵巢、输卵管、子宫中mPRβ表达量较对照组分别降低59.26%、10.25%和8.96%(P>0.05);高脂组在卵巢、输卵管中mPRγ表达量较对照组分别降低10.16%、59.15%(P<0.05),高脂组子宫中mPRγ表达量较对照组降低48.65%(P>0.05)。结果说明营养性肥胖通过对小鼠子宫、卵巢、输卵管体重的影响进而降低孕酮膜受体基因的表达。To probe into the effect of nutritional obesity on the development of mice reproductive organs and the genetic expression of progesterone membrane receptor in the uterus,ovary and fallopian tube.40 female Kunming mice were raised in this test and they were randomly divided into the high-fat group(n=20)and ordinary group(n=20);they were given high-fat diet and ordinary fodder,respectively,in order to construct the model mice with nutritional obesity.After eight weeks of mice fed,the weight of the high-fat group was increased 28.47%compared with the control group(P<0.01),obesity model successfully constructed and collected uterus,ovaries,fallopian tubes were weighed,and then measured mPRα,mPRα,mPRγexpression in the uterus,ovaries,fallopian tubes by quantitative PCR.The results showed that:after 8 weeks,the uterine and ovarian weight of high-fat mice were decreased by 68.39%and 75.27%(P<0.05);the expression of mPRαin the uterus and fallopian tubes of the high-fat group were decreased by 8.65%and 46.50%(P<0.05);the expression of mPRαin the ovary of the high-fat group was decreased by 45.25%(P>0.05);the expression of mPRβin the ovary,fallopian and uterus of the high-fat group were decreased by 59.26%,10.25%and 8.96%(P<0.05);the expression of mPRγin the ovary and fallopian tubes of the high-fat group were decreased by 10.16%and 59.15%(P<0.05),the expreasion of mPRγin the uterus of the high-fat group were decreased by 48.65%(P>0.05).
