摘  要:目的 探讨肥大心肌细胞来源外泌体对RAW 264.7小鼠单核巨噬细胞作用不同的时间后,巨噬细胞的免疫调节作用。方法 肥大心肌细胞模型作为实验组,未经任何处理的心肌细胞作为对照组,实验组和对照组分离纯化的外泌体同时作用RAW 264.7小鼠单核巨噬细胞12 h、24 h、48 h后,检测巨噬细胞分泌的细胞因子IL-6、TNF-α和IL-10的表达趋势。结果 实验组和对照组外泌体在作用巨噬细胞RAW 264.712h、24h后IL-6、TNF-α分泌减少,而IL-10分泌增加,作用48h后细胞因子分泌都减少。结论 肥大心肌细胞来源外泌体通过调节巨噬细胞分泌致炎因子TNF-α、IL-6和相对的抑炎因子IL-10的水平,影响巨噬细胞功能,导致炎症反应发生,推断可能进而影响肥大心肌免疫炎症反应等心肌重塑过程。Objective To investigate the immunomodulatory effects of exosomes derived from hypertrophic cardiomyocytes on RAW264.7 macrophages.Methods The model of hypertrophic cardiomyocyte was used as experimental group and untreated cardiomyocyte as control group.The purified exosomes from experimental group and control group acted on RAW 264.7 mononuclear macrophages for 12,24 and 48 hours at the same time.The expression trends of cytokines IL-6,TNF-alpha and IL-10 secreted by macrophages were detected.Results The secretion of IL-6 and TNF-alpha decreased,while the secretion of IL-10 increased,and the secretion of cytokines decreased after 48 hours.Conclusion The exosomes derived from hypertrophic cardiomyocytes affect the function of macrophages by regulating the levels of TNF-alpha,L-6 and relative anti-inflammatory factor IL-10 secreted by macrophages,leading to inflammation,which may further affect the immuno-inflammatory response of hypertrophic cardiomyocytes and other myocardial remodeling processes.
