半枝莲诱导大肠癌干细胞凋亡

摘　　要：目的观察半枝莲对大肠癌干细胞凋亡的影响,探讨其调控机制。方法运用分选型流式细胞仪分选获得大肠癌干细胞,加入不同浓度的半枝莲(0 mg/mL,0.25 mg/mL,0.5 mg/mL,0.75 mg/mL)处理细胞24 h,采用倒置显微镜观察细胞的形态及密度;通过4%台盼蓝染色检测细胞的增殖情况;运用DAPI染色及流式细胞仪检测细胞凋亡的改变;分别采用Q-PCR、Western blot检测大肠癌干细胞凋亡调控基因Bcl-2、Bax mRNA及蛋白的表达。结果半枝莲可显著抑制大肠癌干细胞的增殖能力(P<0.05),并诱导大肠癌干细胞凋亡(P<0.05),降低Bcl-2/Bax的比值(P<0.05)。结论半枝莲具有促进大肠癌干细胞凋亡的作用,通过影响Bcl-2及Bax两者之间的平衡是其可能的调控机制之一。Objective:To study whether Scutellaria Barbata D.Don induce the apoptosis of colorectal cancer stem cells and explore its regulatory molecular mechanism.Methods:Colorectal cancer stem cells were obtained from SW480 cell line by flow cytom etry and treated with Scutellaria Barbata D.Don(0 mg/mL,0.25 mg/mL,0.5 mg/mL,0.75 mg/mL) for 24 h.The effect of Scutellaria Barbata D.Don on colorectal cancer stem cells proliferation was detected by inverted microscope and cell counter with 4% trypan blue staining.DAPI and Annexin V-APC/PI assay were used to observe the effect of Scutellaria Barbata D.Don on the apoptosis of colorectal cancer stem cells.Q-PCR and Western Blot were used to detect the expression of Bcl-2 and Bax in colorectal cancer stem cells.Results:These results showed that Scutellaria Barbata D.Don could significantly inhibit the proliferation(P<0.05) and induce the apoptosis of colorectal cancer stem cells,while decrease the ratio of Bcl-2/Bax(P<0.05).Conclusion:Scutellaria Barbata D.Don could induce the apoptosis of colorectal cancer stem cells by a possible way of decreasing the ratio of Bcl-2/Bax.
