摘  要:以功能性离子液体作为桥联剂,将天然有机高分子材料壳聚糖(CS)和介孔SiO2(SBA-15)制备一种新型有机-无机纳米复合材料。以合成的复合材料为载体,用于猪胰脂肪酶(PPL)的固定化,并在有机相中对其催化α-苯乙醇转酯化拆分反应进行研究。结果表明:新型纳米复合材料固定化PPL在最优工艺条件(反应温度45℃、反应时间13 h、醋酸乙烯酯和苯乙醇的摩尔比8∶1、底物浓度3 mol/L、转速200 r/min)下,对消旋α-苯乙醇与乙酸乙烯酯的转酯化拆分活性和立体选择性有了大幅度提升。在转化率为55.3%时,底物S-对映体过剩值可达99.3%,对映体选择性可达77.2%,相比游离酶提高了13.04倍。研究证明基于功能性离子液体连接的壳聚糖-介孔硅纳米材料是提高酶选择性和催化效率的良好载体。A novel organic-inorganic nanocomposite was prepared by using natural organic polymer chitosan(CS) and mesoporous SiO2(SBA-15) as components and functional ionic liquids as bridging agent. The composite material was used as carrier for immobilization of porcine pancreatic lipase(PPL) and its catalytic transesterification and resolution of α-phenylethanol in organic phase was investigated. Results showed that the transesterification activity and stereoselectivity of racemic α-phenylethanol and vinyl acetate were greatly improved under the optimized conditions of reaction temperature of 45 ℃, reaction time of 13 h, raw material molar ratio of vinyl acetate to phenylethanol of 8∶1, substrate concentration of 3 mol/L, and rotation speed of 200 r/min. When the conversion rate was 55.3%, the substrate S-enantiomer excess value was 99.3%, and the enantiomer selectivity was 77.2%, which was 13.04 times higher than that of free enzyme. It was proved that chitosan-mesoporous silica nanomaterials based on functional ionic liquids were good carriers for improving enzyme selectivity and catalytic efficiency.
