护卵汤对卵巢储备功能减退大鼠卵巢指数及卵泡数量的影响

摘　　要：目的:观察中药护卵汤对卵巢储备功能减退(DOR)大鼠卵巢指数及卵泡数量的影响。方法:选取动情周期为4?5d的雌性大鼠48只为实验动物,随机数字表法,将其分为空白组、模型组、护卵汤组和补佳乐组4组,每组12只。采用环磷酰胺(CTX)单次腹腔注射的方法制备DOR大鼠模型。自造模第1天开始,每天给予护卵汤组大鼠1 mL/100 g护卵汤浓缩液灌胃;每天给予补佳乐组大鼠1 mL/100 g补佳乐稀释液灌胃;空白组、模型组大鼠每天按相同剂量给予生理盐水灌胃。每天1次,连续4周,每周休息1d。给药4周后称取卵巢湿重并计算卵巢指数,观察卵巢组织形态学变化,累积计数原始卵泡、初级卵泡、窦状卵泡和闭锁卵泡数目。结果:与空白组相比,模型组大鼠双侧卵巢湿重、卵巢指数均降低(P<0.01),卵巢组织损伤明显,原始卵泡、初级卵泡、窦状卵泡数量均减少(P<0.05,P<0.01),闭锁卵泡数量增加(P<0.01);与模型组相比,护卵汤组和补佳乐组大鼠双侧卵巢湿重、卵巢指数均增高(P<0.01,P<0.05),卵巢组织损伤较轻,护卵汤组大鼠卵巢组织中原始卵泡、初级卵泡数量均增加(P<0.05,P<0.01),闭锁卵泡数量减少(P<0.05);补佳乐组大鼠卵巢组织中初级卵泡数量增加(P<0.05),闭锁卵泡数量减少(P<0.05)。结论:护卵汤具有改善CTX所致DOR大鼠模型的卵巢指数,减轻卵巢组织损伤,提高卵泡数量,从而保护卵巢功能的作用。Objective:To observe the effects of Huluan decoction on ovary index and the number of follicles in rats with Diminished Ovarian Reserve(DOR). Method:48 female rats with an estrous cycle of 4-5 days were selected as experimental animals.The rats were randomly divided into four groups,the normal group,the model group,the Huluan decoction group and the Bujiale group,with 12 rats in each group. The animal model of rats with DOR was established by single intraperitoneal injection of cyclophosphamide. The Huluan decoction group and the Bujiale group were given corresponding liquid(1 mL/100 g)gavage,the blank group and model group were given normal saline(1 mL/100 g)gavage,once daily for 4 weeks from the first day of model establishment.After the end of 4 weeks of administration,ovarian wet weight was weighed and the ovarian index was calculated,morphological changes of ovarian tissue were observed,the number of primordial follicles,primary follicles,sinusoid follicles and atresia follicles were counted. Result:Compared with the normal group,the ovarian wet weight and ovarian index of the model group were decreased(P<0.01),the ovarian tissue damage was obvious,and the number of primordial follicles,primary follicles and sinus follicles were significantly decreased(P<0.05,P<0.05),the number of occluded follicles increased significantly(P<0.01). Compared with the model group,the ovarian wet weight and ovarian index of the rats in the Huluan decoction group and the Bujiale group increased(P<0.01,P <0.05),the ovarian tissue damage was lighter,the number of primordial follicles and primary follicles increased significantly(P<0.05,P<0.01)and the number of atretic follicles decreased significantly(P<0.05)in the ovarian tissue of the rats in the Huluan decoction group.The number of primary follicles was significantly increased(P<0.05)and the number of atretic follicles was significantly decreased(P <0.05)in the ovarian tissue of the rats in the Bujiale group.
