摘 要:蜕皮激素20E调控昆虫的蜕皮与变态,E74是昆虫蜕皮激素信号传导过程中关键转录因子之一。以飞蝗(Locusta migratoria)为研究对象,对LmE74A基因的表达、调控及功能进行了研究。搜索飞蝗转录组数据库获得E74A基因全长cDNA序列,命名为LmE74A(GenBank登录号:MK568625),其中开放阅读框(ORF)长度为1539bp,编码512个氨基酸。Reverse transcription-quantitative PCR(RT-qPCR)分析表明:LmE74A在飞蝗体壁中高表达,其次是前肠和后肠;LmE74A在五龄第5天表达量最高,蜕皮前后表达量较低。体内注射20E后,LmE74A的表达显著上升,说明LmE74A可以被20E诱导。在五龄期注射LmE74A的dsRNA后,注射dsLmE74A组的LmE74A基因的表达量较对照组显著降低,但该基因的表达下调并没有影响飞蝗正常羽化为成虫。The molting and metamorphosis of insects are regulated by ecdysone hormone 20Eand E74is one of the key transcription factor in the signaling pathway of 20E.The expression,regulation and function of E74Agene in Locusta migratoria were studied.By searching the locust transcriptome database,the fulllength cDNA sequence of E74A gene was obtained,named LmE74A(GenBank accession number MK568625).It contained a 1539-nucleotide open reading frame(ORF)encoding 512amino acid residues.RT-qPCR analysis showed LmE74A was predominantly expressed in the integument,followed by foregut and hindgut.The expression of LmE74Areached peak on the fifth day of the 5thinstar nymphs,and relatively lowly expressed before and after molting.The expression of LmE74A was significantly increased after the injection of 20Einto locusts,which suggested that LmE74Acould be induced by 20E.When dsRNA of LmE74A was injected into the 5thinstar nymphs,the expression of LmE74A was greatly repressed in dsLmE74A-injected insects compared to controls.However,the down-regulated expression of LmE74A did not affect locusts’molt into adults successfully.
